اين ژن در تعدادي از جدايههاي موجود در بانك ژن به كمك نرم افزار 7 89 CLC Sequence Viewer شكل3 42 قطعه 039جفت بازي تكثير يافته ژن nodC با استفاده از جفت آغازگر اختصاصي nodCI nodCF در جدايههاي ريزوبيومي منتخب همزيست با اسپرس 09 شكل3 52 قطعه087جفت بازي تكثير يافته ژن nifH با استفاده از جفت آغازگر اختصاصي 2 nifH 1 nifH در جدايههاي ريزوبيومي منتخب همزيست با اسپرس 29 شكل3 62 تشكيل گره بر روي ريشه گياه اسپرس از گونه O viciifolia Scop توده نجفآباد و O arnaria تلقيح شده با دو جدايه 4 ROsnA و 5 ROpA بصورت مجزا و توأمان در ماسه استريل 79 پانز چكيده ريزوبيومها از جمله مهمترين باكتريهاي خاكزياند كه با ايجاد يك اختصاصي با گياهان خانواده لگومينوز به تثبيت رابطه همزيستي كاملا نيتروژن مولكولي ميپردازند برقراري رابطه همزيستي ريزوبيوم لگوم مستلزم تبادل يكسري از سيگنالهاي بيوشيميايي از دو طرف رابطه منجر ميشود از همزيستي ميباشد كه به شناسايي آنها توسط يكديگر آنجايي كه خصوصيات ژنتيكي ريزوبيومهاي همزيست و ميزان سازگاري آنها با گياهان لگوم ميزبان مهمترين عامل تعيينكننده رقابت نژادهاي مختلف براي گرهزايي ميباشد اطلاع از تنوع ميان جمعيت نژادهاي موجود

Identification of Sainfoin Species Nodulating Rhizobia Morteza Ghorbani Morteza Ghorbani@ag iut ac ir January 12 2015 Department of Agricultural Biotechnology Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language FarsiSupervisors M Talebi mtalebi@cc iut ac ir M Bahar mbahar@cc iut ac irAbstractRhizobiums is one of the most important bacteria in the soil The symbiotic intraction between the rhizobia andleguminous plants is a highly sophisticated process with exchange of biochemical signals between two partnerswhich leads to molecular nitrogen fixation Genetic characterization of symbiotic rhizobium and its compatibilitywith the host legume is the most important factor to determinant the nodulation competitiveness of different strains therefore identification of genetic diversity among the different rhizobia in the soils the frequency of strains anddetection of compatible strains with different host species is very important Sainfoin due to desirable features suchas high nutritional value lack of bloat in cattle maintain soil fertility resistance to drought salinity cold and pestssuch as Hypera postica was cultivation especially is regions where there is no possibility of alfalfa and clovercultivation The symbiont sainfoin rhizobiums have shown a high heterogeneity and the relationships betweensymbiotic rhizobium strains with host species related to geographic region is unknown The West of Isfahanprovince Friedan and Fereydoonshahr and neighboring regions of Lorestan province Azna and Aligudarz areimportant areas of sainfoin cultivation and it is essential to identify symbiotic rhizobium species nodulating sainfoinin these regions Therefore in this study classification of symbiotic strains with different species of sainfoin 7species and 4 populations based on morphological and molecular PCR RFLP of the 16S rDNA genomic region methods grouped the isolates in four groups In order to increase the accuracy of grouping and evaluation of geneticdiversity representatives of groups were selected and more studied using rep PCR fingerprinting BOX A R andERIC primers and the results confirmed the previous classification Sequencing of 16S rDNA genomic region inselected isolates showed that the most isolates nodulating sainfoin species in both Isfahan and Lorestan soilbelonged to Rhizobium gallicum and Sinorhizobium meliloti species with visible dominance of R gallicum Nodulating of S meliloti with sainfoin was reported for first time In this study Agrobacterium tumefaciens isolateswere also observed in sainfoin nodules Detection of effective genes in symbiosis nifH and nodC revealed theexistence of nodding gene nodC in the isolates but nitrogen fixation gene nifH was absent in A tumefaciens Theability of re nodulating and competition between the two strains of R gallicum and S meliloti species wasexamined and the results confirmed the nodulation of both species with priority of R gallicum Melanin productionassay cannot detect any positive in all of 236 isolates Detection of R gallicum as symbiotic species with sainfoin according to previous studies was expected and its predominance in competition with S meliloti can be due to morespecificity of this symbiosis It seems that the inhibitory effect of A tumefaciens on R gallicum strain in natural soilcan be decrease dominance of R gallicum in symbiosis with sainfoin Keywords Rhizobium molecular classification Sainfoin competition Melanin
