93 A comparative evaluation of osteoinductivity and antibacterial effects of the bioactive glass and hydroxyapatite nanoparticles in hard tissue engineering AmirHossein Tavakolizadeh a tavakolizadeh@ma iut ac ir Date of Submission June 24 2015 Department of Materials Engineering Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Language FarsiSupervisors Mahdi Ahmadian ahmadian@cc iut ac ir Mohammadhossein Fathi fathi@cc iut ac ir AbstractBecause of biocompatibility and desirable bioactivity bioceramics are the most suitablematerials for the replacement or repair of damaged hard tissue of the body treatment of diseaseand disability and even replacement of the soft tissues of the human body Also in recent years because of biological properties high bioactivity and osteoinduction properties bioactive glass bioglass have been favored On the other hand the application of Hydroxyapatite HA mineral phase of the bone has been expanding in types of natural and synthetic kinds Thisstudy aimed at preparation osteoinduction evaluation and antibacterial evaluation ofnanobioglass and hydroxyapatite nanoparticles 63S 58S and 45S bioactive glasses and HAnanoparticles were obtained by sol gel process Bioceramic specimens were characterized byX ray diffraction XRD scanning electron microscopy SEM energy dispersive X rayanalysis EDX fourier transform infrared spectroscopy FTIR and laser dopplerelectrophoresis LDE As an index of bioactivity HA formation ability on bioceramicspecimens was evaluated in simulated body fluid SBF The capacity of prepared nanoparticlesfor bone formation was investigated in vitro using human cord blood derived unrestrictedsomatic stem cells USSC under osteogenic induction and toxicology of specimens was carriedout by conventional MTT method Antibacterial activity of the samples was also evaluated Thezeta potential was negative for all compositions in distilled water The results showed that invitro biocompatibility and bioactivity of bioactive glasses are higher than hydroxyapatite These findings confirmed by SBF and toxicology tests During osteogenic differentiation significantly higher values of ALP activity and biomineralization were observed onhydroxyapatite and 45S bioactive glass compared to 58S and 63S All nanopowders showedantibacterial effects on E coli and S aureus while hydroxyapatite and 45S bioactive glassdemonstrated better effects compared to 58S and 63S Key words Bioactive glass Hydroxyapatite Osteoinduction Antibacterial activity
