1 Preparation and evaluation of meloxicam nanoparticles in an injectable hydrogel matrix for cartilage regeneration Shohreh Fattahpour Biomaterials Research Group Department of Materials Engineering Isfahan University of Technology Isfahan 841456 83111 Iran sh fattahpour@gmail com July 2015 Prof Morteza Shamanian shamanian@cc iut ac ir 1 supervisor Dr Naser Tavakoli tavakoli@pharm mui ac ir 2 supervisorAdvisor Prof Mohammadhossein FathiAdvisor Prof Hojjat SadeghiDepartment graduate coordinator Dr Keyvan RaeissiAbstractRecently tissue engineering is common for regeneration of damages in this way scaffoldsare important for protecting of extra cellular matrix Hydrogel scaffolds are unique incartilage drug delivery and tissue engineering The basic aim of this research wasdevelopment of a novel methyl cellulose MC carboxymethyl chitosan CMC pluronicand zinc hydrogel with meloxicam nanoparticles NPs NPs were designed by Box behnken analysis according to alginate chitosan mass ratio pluronic and meloxicamconcentration as variables and particle size zeta potential encapsulation efficiency EE dissolution efficiency DE and mean dissolution time MDT as dependent responses Theoptimized formulation was prepared based on the lower particle size and the higher EEcriteria CMC a water soluble derivative of chitosan was processed then MC and CMChydrogels were prepared Gelation time and gelation temperature of hydrogels weredetermined NPs were added to the hydrogel base for long remaining in articular cartilageand local drug delivery Pluronic solution was used for proper NPs dispersion andavoidance of agglomeration The hydrogels degradation morphology ATR FTIR spectra rheology drug release and cell viability was investigated The results showed that althoughalginate chitosan mass ratio pluronic and meloxicam concentrations were affected onparticle size the effect of alginate chitosan mass ratio is more this factor significantlyimpressed on zeta potential EE DE and MDT were affected by meloxicam concentration The hydrogel gelation time was increased by MC and CMC ratios Swelling was increased
