109 Changes in the Cell Wall Composition and Physiological Properties of Alfalfa in Response to Toluene Contamination and Silicon Nutrition Masume Sharifi sharifimsm@yahoo com May 14 2016 Department of Soil Science College of Agriculture Isfahan University of Technology Isfahan 84156 83111 Iran1st Supervisor A H Khoshgoftarmanesh amirhkhosh@cc iut ac ir2nd Advisor F Ghanati ghangia@modares ac ir3rd Advisor H Hadadzadeh hadad@cc iut ac ir4th Advisor M H Ehtemam hehtemam@cc iut ac irCollege Graduate Coordinator M M Majidi1 Professor Department of Soil Science College of Agriculture Isfahan University of Technology Isfahan 84156 83111 Iran 2 Associate Professor Department of Plant Biology Faculty of Biological Sciences Tarbiat Modares University Tehran Iran3 Professor Department of Chemistry Isfahan University of Technology Isfahan 84156 83111 Iran4 Assistant Professor Department of Agronomy and Plant Breeding Collage of Agriculture Isfahan University of Technology 84156 83111 Iran AbstractSilicon Si is a beneficial nutrient that its role on cell wall composition and improvement of plant toleranceto environmental stress has been widely reported Toluene is an aromatic petroleum hydrocarbon withharmful effects on plants Therefore to understand the mechanisms by which Si nutrition affects planttolerance to toluene contamination knowledge on the changes in the root CWs is needed To our knowledge there is a lack of information on the changes in plant root cell wall in response to toluene contamination andSi nutrition In the present study the effects of Si nutrition 0 and 1 mM on the CW components andproperties and also selected physiological responses of the alfalfa Medicago sativa L plants exposed totoluene 0 225 and 450 mg L 1 were examined in the nutrient solution culture Our results indicated thattoluene contamination induced an oxidative stress and significantly reduced the growth of alfalfa Exposureto toluene significantly decreased the ion exchange capacity CEC and swelling coefficient Kcw of theCWs by a reduction in the total number of the root CWs functional groups The exposure to toluenesignificantly decreased the alfalfa root CW dry mass and modified distribution of polysaccharides in the CW Toluene exposure enhanced activity of lignifying peroxidases and lignin content of the alfalfa root CW These changes were accompanied with reduced water potential of alfalfa leaves Supplement with Si led to asignificant increase in the alfalfa root fresh mass particularly in the plants exposed to toluene Addition of Sialso reduced toluene induced changes in distribution of polysaccharides and lignin content of alfalfa root
