Genetic Assessment of Captive bred Wild sheep in Chadegan Recreation Village Naeem Kaveh Pishghadam naeem kaveh@gmail com July 1 2016 Department of Natural Resources Engineering Isfahan University of Technology Isfahan 84156 83111 IranDegree MSc Language PersianSupervisor Dr Mansoureh Malekian mmalekian@cc iut ac irAbstractLosses of biodiversity and species extinction are considered as global crises which has initiatedconservation efforts to protect species and reduce the extinction rate Captive breeding is one of theways to increase the populations of a species and to re introduce them to the wild Captive breedingprojects are time consuming costly and might fail due to genetic problems such as inbreeding Therefore genetic studies are prerequisites for such projects The current study aimed to determinethe taxonomic status and phylogenetic relationship of the sheep population in Chadegan recreationvillage to comprehend the origin and potential sites in the case of releasing animals to the wild andto assess the genetic diversity of the current population DNA extraction from placental tissuesamples from six individuals was performed using ammonium acetate method PCR reactions wereperformed using specific cytochrome b primers and a fragment of 688 base pairs bp wasamplified and sequenced Available sequences of Iranian sheep populations were extracted fromGenebank and used for phylogenetic reconstruction Results showed that Iranian wild sheep couldbe divided into two Clades namely O orientalis and O vignei with posterior probability of 0 99 and98 9 bootstrap support Based on this classification the captive bred population of Chadeganrecreation village belonged to O vignei and the subspecies of O vignei laristanica with highposterior probability and bootstrap support Evaluating the genetic diversity of wild sheep based onthe 688 bp sequenced fragment revealed 67 haplotypes among the 133 available sequences Threenew haplotypes were identified in the current study and added to the previous 67 haplotypes whichshow a relatively high genetic variation in Chadegan recreation village population The geneticdivergence within and between O vignei O orientalis O canadensis and O ammon wascalculated using K2P method Minimum and maximum intra specific genetic distance within O vignei were obtained from 0 013 to 0 015 while the inter specific genetic distance between O vignei and O canadensis ranged from 0 086 to 0 088 and the distance between O vignei and O orientalis varied from 0 021 to 0 24 The lowest intra species genetic distance was observedbetween subspecies of O canadensis and O orientalis while the maximum value was obtained inO vignei subspecies Considering the genetic distances that exist between the subspecies transferring individuals between populations should be performed with adequate care to preservethe intra specific diversity of wild sheep Keywords captive breeding Chadegan cytochrome b genetic diversity phylogeny wild sheep
