Identification of male sterile maintainer lines in onion using PCR based methods Raheleh Dehghani r dehghani@ag iut ac ir September 22 2018 Department of Agronomy and Plant Breeding Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language Farsi1st Supervisor Majid Talebi mtalebi@cc iut ac ir2nd Supervisor Badraldinebrahim Sayed Tabatabaei sayedt@cc iut ac irAbstractOnion Allium cepa L is one of the most important vegetables cultivated for its widespread consumptionof food and medicine Onion is a highly cross pollinated crop that lead to non uniformity morphology genetics low quality and performance of seeds The production of hybrid seeds of vegetables is a majornecessity in agriculture Hybrid seed production in onion involves the use of male sterility In the hybridseed production project prepare and reproduce three line is essential that A lines with the genotype S msms as maternal parent and B with genotype N msms as a maintainer parent for A line and C as apaternal parent with active and fertile pollen that maintainer lines is needed to multiplication of male sterileline Onion is a biennial there for identification of the maintainer line using traditional and classic methodsof cross test require at least four years nowadays using of biotechnology and molecular markers it ispossible to accelerate identification the maintainer lines and the cytoplasm S was identified from N as wellas Locus ms at a very short time Therefore this project was conducted with the aim of evaluating theeffectiveness of molecular markers in identifying the type of cytoplasm and diagnosing the genetic malesterility factors in the nucleus In this study PCR STS molecular markers that were introduced and verifedby various researchers The three cytoplasmic molecular markers cob accD MK were used to identifythe cytoplasm S from the normal cytoplasm N and to identify the type of male sterile cytoplasm S fromT type and the four nuclear molecular markers OPT PsaO Jnurf 13 and AcSKP1 were used to detectthe genetic male sterility factors in the nucleus based on the length and number of amplified fragments The plant material used 123 samples consisted of male sterile maintainer and backcross lines externaland internal hybrids and open pollinated populations The efficiency of cytoplasmic markers was firstinvestigated on the male sterile backcross and maintainer lines and then on populations results showedthat the accD and cob markers in detection of normal and S type of cytoplasm were quite similar and showed100 efficiency In addition to identify the type of normal cytoplasm from the male strile type S MKmarker was able to distinguish between two types of S from T with a 100 efficiency The use of fournuclear markers was not successful in genotype detection and locus ms The main emphasis was oncytoplasmic markers in order to identify the male sterile cytoplasms remove them from the population andreduce the number of cross linked genetic material all of which were performed with 100 efficiency Key wordsAllium cepa Cytoplasmic male sterility Maintainer line Cytoplasmic markers Nuclear markers
