121 AbstractThe aim of this thesis is the investigation of drug release potential of poly vinyl alcohol PVA chitosan CS polyhedral oligomeric silsesquioxane POSS nanocompositehydrogels which is crosslinked by gamma irradiation Regarding this goal the effect of type amount of nanoparticles and irradiation dose on the microstructure properties and drugrelease of the nanocomposite was studied A series of casted films from aqueous solution ofPVA CS POSS were prepared Weight ratio of PVA CS kept constant 90 10 and variouscontents of hydrophilic POSS 1 3 and 5 wt and amphiphilic POSS 0 5 1 5 and 2 5 wt were used respectively The films were dried irradiated at doses 20 40 kGy followedby annealing in an oven In the first step the effects of POSS contents and irradiation doseson physical properties of all samples were investigated The result indicated that at all doses the gel content and the tensile strength of the hydrogels decreased while swelling andelongation at break increased with increase in hydrophilic nanoparticle content Water vaportransition rate WVTR of the samples improved with hydrophilic POSS incorporation anddecreased with increase in irradiation dose The gel content increased as the amount ofamphiphilic nanoparticles increased up to 1 5 wt followed by a decrease at 2 5wt Thewater absorption tensile strength and elongation at break decreased with increasing amountof amphiphilic POSS At 0 5 and 1 5 wt of amphiphilic nanoparticle at all irradiationdoses the WVTR of the samples was enhanced and decreased at 2 5wt In the secondstep samples with the suitable physical properties for wound dressing application wereselected characterized and their release of gentamicin sulfate was investigated Drugrelease profiles from hydrophilic nanocomposite hydrogel can be divided into three stages the burst release during the initial 3 4h period the increasing release rate during the 3 12 h period and the nearly constant release rate during the 12 24 h period The drug releaseprofile obtained for the amphiphilc nanocomposites showed a burst effect followedby a relatively slow release A Fickian diffusion mechanism was found for the drug releasefrom hydrophilic and amphiphic samples The samples contained 3 wt of hydrophilicPOSS or 0 5 wt of amphiphilic POSS and irradiated at 30kGy were cytocompatible andexhibited antibacterial activity Key words Polyvinyl alcohol Nanocomposite hydrogel Drug delivery POSS Gammairradiation
