Comparison of Secondary Metabolites in Dorema ammoniacum D Don and Dorema aucheri Boiss Somayeh Arabjafari arabjafarisomayeh@gmail com January 2021 Department of Natural Resources Isfahan University of Technology Isfahan 84156 83111 IranDegree MSc Language PersianDr P Golkar golkar@cc iut ac irDr M Tarkesh Esfahani m tarkesh@cc iut ac irAbstractThe Umbelliferae family is one of the most important medicinal plant families and due to its secondarymetabolites such as essential oils and antioxidants it has a wide distribution in different climatic regions The genus Dorema has six species and belongs to the Umbelliferae family of which two species Vosha Dorema ammoniacum D Don and Bilher Dorema aucheri Boiss are native species of Iran The aimof this study was to investigate and compare the composition of essential oils and secondary metabolitesin the leaf and stem organs of two species Vosha and Bilhar from 14 different habitats For this purpose shoots including leaves and stems were collected and completely dried and then ground separately Then the essential oil was extracted by steam distillation by Clevenger and analyzed by gas chromatography mass spectrometry The compounds in the essential oil belong to 5 main chemical groups including hydrocarbon monoterpene oxygen monoterpene sesquiterpene hydrocarbon oxygen diterpene andhydrocarbons in different genes of Vosha and Bilher were determined that among them hydrocarbonsesquiterpenes had the highest amount of essential oil compounds in all genotypes The major compoundsidentified in Biller essential oil were Caryophylline Beta Calaren Tetradcan Alpha Himachallen andLinalool These compounds are used in the pharmaceutical and cosmetic industries The number ofidentified compounds in Biller genotypes was more than Vosha genotypes Two genotypes of Bilhar Shabliz Kohgiluyeh and Boyerahmad and Islamabad Fereydunshahr had the highest essential oil yieldwith 0 39 and 0 36 respectively In the second part of this study extracts of leaf and stem organswere performed with 80 methanol Total phenol content was determined using the Folin Cicaltomethod total flavonoid content and flavonol were determined by aluminum chloride staining method Antioxidant activity was also determined using DPPH method The results of mean comparison showedthat in all genotypes the amount of active ingredients and antioxidant properties of leaves were higherthan stems Based on the comparison of the average biochemical traits of Biller in different genotypes the highest amount of total phenol content 34 3 mg gallic acid dry weight and antioxidant activity 92 were identified in Margoon Kohgiluyeh and Boyer Ahmad genotypes The highest content of totalflavonoids 5 5 mg quercetin dry weight was observed in Ardal Chaharmahal and Bakhtiari genotype In two genotypes the highest content of total flavonoids 4 2 mg quercetin dry weight total phenol 24 5 mg gallic acid dry weight total flavonols 2 9 mg quercetin weight Dry and antioxidant activity 80 9 were identified in Fereydunshahr Bardasiab genotype The difference in the amount of secondarymetabolites of the studied genotypes may be due to differences in habitat and climatic conditions Asignificant positive correlation was observed between the amount of total phenol and the amount of totalflavonoids in both species There was also a significant positive correlation between antioxidant activityand total flavonoid content Due to the positive correlation between the total flavonoid content andantioxidants in both leaf samples it seems that in these species the antioxidant activity is related to thetotal flavonoid content and flavonoids have antioxidant properties Keywords Essential oil Antioxidant activity Bilhar Total phenol content Total flavonoid content Vosha
