Therefore, in this study, with the aim of isolating Bifidobacteria from 2 samples of local yogurt and 30 samples of feces of nomadic livestock such as cows, donkeys, buffaloes and goats were sampled.The samples were quickly collected in clean containers and stored with ice packs and immediately transferred to the laboratory. First, the sample was homogenized and after weighing, it was diluted by cystine-containing peptone buffer and It was cultivated linearly from all dilutions on Bearins and TPY media. then stored under anaerobic conditions controlled with 10% carbon dioxide at 37 ° C for 3-5 days.After the growth of the colonies, morphological tests were performed to identify them, such as examining the appearance of the colonies, simple staining, and observation under a microscope to examine the arrangement and cell shape of the microbes. Biochemical tests such as Gram staining, spore staining, catalase test, nitrate reduction, Indole production from tryptophan, aerobic test, production of gas from glucose, carbohydrate fermentation test such as salicin, lactose, mannitol, ribose, gluconate, trehalose , Melibios, Maltose, Arabinose, Xylose, Rafinose, Sucrose, Mannose, Cellobiose, Starch, Inulin, Fructose, And the most key test of Bifidobacter family was the presence of fructose-6-phosphate phosphoctolase on pure culture of bacterial isolates.During these experiments, of 89 isolates, 32 isolates of Bacillus and 19 isolates with different abundance in different species of Bifidobacteriu Such as Bifidobacterium catenulatum, Bifidobacterium gallinarum, Bifidobacterium breve, Bifidobacterium scardovii, Bifidobacterium thermophilum, Bifidobacterium thermacidophilum, Bifidobacterium adolescentis, Bifidobacterium animalis, And 7 isolates of Bifidobacterium with different fermentation patterns were identified, which are expected to be new strains of .BifidobacteriumFinally, after performing a molecular test with specific primers, the results of the research were confirmed.During this study, it was found that feces of native animals are rich sources of probiotics. Which can be studied as a powerful source of extraction of probiotic strains, And with their help, a good collection of probiotics can be collected.
