تغييرات اقليمي، اثرات گلوگاه ژنتيكي (Genetic bottleneck) ناشي از هزاران سال اهليسازي و انتخاب مصنوعي را آشكارتر كرده است؛ محدوديتي كه توسعه ارقام سازگار با تنشهاي همزمان خشكي، گرما و بهويژه شوري را در مناطق خشك و نيمهخشك دشوار ساخته. شوري خاك با اختلال همزمان در روابط آبي، هموستازي يوني، تنظيم اسمزي و تعادل اكسيداتيو، تحمل گياه به آن را به صفتي پيچيده و چندبعدي تبديل ميكند. بنابراين بازگشت هدفمند به ذخاير ژنتيكي جو وحشي اسپانتانئوم (Hordeum spontaneum) ميتواند راهبردي مؤثر براي گسترش پايه ژنتيكي جو زراعي (Hordeum vulgare) و توسعه ژنوتيپهاي سازگار با شوري باشد. اين پژوهش با هدف بررسي تنوع و تركيبهاي ژنتيكي حاصل از ادغام آشيانهاي ژنوم جو وحشي در زمينه ژنتيكي جو زراعي، در جمعيت نوتركيبي كه براي نخستين بار در ايران از تلاقي ژرمپلاسم وحشي جو با تأكيد بر تنوع ژنتيكي خويشاوندان وحشي فلات ايران ايجاد شد، شناسايي منابع ژنتيكي و لاينهاي برتر براي تحمل شوري، و تبيين سازوكارهاي فيزيولوژيك مرتبط، در قالب دو مطالعه انجام شد. در مطالعه نخست، جمعيت آشيانهاي IWB-21 (443 لاين اينبرد نوتركيب حاصل از تلاقي 21 ژنوتيپ جو وحشي با رقم ريحان 03) از نظر صفات زراعي و مورفولوژيك براي كل جمعيت، و صفات فيزيولوژيك، يوني و بيوشيميايي براي 85 ژنوتيپ منتخب، در شرايط نرمال و شور طي دو سال زراعي ارزيابي شد. در مطالعه دوم، 121 ژنوتيپ شامل ارقام زراعي، اكسشنهاي بانك ژن، لاينهاي نوتركيب منتخب از جمعيت آشيانهاي قبلي و ژنوتيپهاي جو وحشي اسپانتانئوم در دو شرايط نرمال و شور و طي دو سال زراعي از نظر صفات مرفولوژيك، فيزيولوژيك، يوني و بيوشيميايي ارزيابي شدند. تاريخ كشت ژنوتيپها در هر دو مطالعه با هدف بازتاب شرايط واقعي كشت تاخيري در منطقه، 22 بهمنماه در دو سال زراعي 1401-1400 و 1402-1401 در نظر گرفته شد. نتايج مطالعه نخست نشان داد شوري اكثر صفات زراعي از جمله عملكرد دانه، عملكرد بيولوژيك و شاخص برداشت را بهطور معنيداري كاهش داد، اما تنوع فنوتيپي گسترده در جمعيت IWB-21 ظرفيت بالاي آن را براي شناسايي تركيبهاي ژنتيكي متفاوت و سازگار با شرايط تنش نشان داد. در اين ميان، آثار ادغام بخشهاي ژنومي نمونههاي متفاوت وحشي در پسزمينه ژنتيكي ارقام زراعي و نقش اين فرآيند در شكلگيري تركيبهاي فنوتيپي متنوع و قابل استفاده براي اهداف مختلف اصلاحي، بهوضوح مشهود بود. شواهد تفكيك متجاوز حاكي از آن بود كه بازتركيبي آللهاي وحشي و زراعي ميتواند به تركيبهاي جديدي از ژنوتيپها با عملكرد و ويژگيهاي مطلوبتر از والدين منجر شود؛ خانوادههاي 10، 13 و 15 و لاينهاي 257RIL، 283RIL و 405RIL بهعنوان ژنوتيپهاي برتر از نظر تركيب عملكرد و صفات مرتبط با تحمل به شوري شناسايي شدند. از نظر فيزيولوژيك، شوري با كاهش محتواي آب نسبي و پتاسيم و افزايش پرولين، سديم، نسبت سديم به پتاسيم و فعاليت آنزيمهاي آنتياكسيداني همراه بود، اما لاينهاي برتر بيشتر از طريق تعادل ميان عملكرد و پاسخ فيزيولوژيك سازگاري نشان دادند تا پاسخ دفاعي شديد. در مطالعه دوم نيز تنوع ژنتيكي گستردهاي ميان گروههاي مختلف ژرمپلاسم مشاهده شد. لاينهاي نوتركيب با ميانگين عملكرد حدود 489 گرم بر متر مربع در شرايط شور، كمترين كاهش عملكرد (حدود 26٪) و بهترين جايگاه از نظر شاخصهاي تحمل را داشتند. در ميان ژنوتيپهاي برتر، 180RIL، 287RIL، 427RIL، 247 RIL و281RIL بيشترين ارزش را بر اساس تركيب عملكرد و شاخص تركيبي معنيدار (CSI) نشان دادند؛ 287 RIL با تنها حدود 3٪ كاهش عملكرد، نمونهاي برجسته از پايداري بود. شاخص تركيبي معنيدار (CSI) نسبت به شاخص پايداري عملكرد (YSI) ارتباط قويتري با عملكرد واقعي تحت شوري نشان داد. عملكرد دانه در هر دو محيط وراثتپذيري بالايي داشت و تعداد سنبله در متر مربع بهعنوان معيار غيرمستقيم مناسب براي انتخاب تحت تنش شناسايي شد. در مجموع، نتايج دو مطالعه نشان داد كه تنوع ژنتيكي جو وحشي، بهويژه هنگامي كه از طريق جمعيتهاي آشيانهاي و لاينهاي نوتركيب به زمينه ژنتيكي زراعي منتقل و بازتركيب ميشود، ظرفيت قابلتوجهي براي ايجاد تركيبهاي جديد و مطلوب عملكرد و تحمل به شوري دارد. يافتههاي پژوهش همچنين نشان داد كه ارزيابي تحمل به شوري بايد بر پايه تركيبي از عملكرد دانه، صفات فيزيولوژيك، تعادل يوني و شاخصهاي چندصفتي انجام شود. لاينهاي برتر شناساييشده در اين پژوهش ميتوانند بهعنوان منابع ارزشمند در مطالعات ژنتيكي، پيشبهنژادي و برنامههاي اصلاحي با هدف توسعه ژنوتيپهاي سازگارتر با شرايط شور مورد استفاده قرار گيرند.

Climate change is increasingly revealing the effects of a genetic bottleneck resulting from thousands of years of domestication and artificial selection—a constraint that has made the development of cultivars adapted to simultaneous drought, heat, and especially salinity stress in arid and semi-arid regions increasingly difficult. Soil salinity, by simultaneously disrupting water relations, ionic homeostasis, osmotic regulation, and oxidative balance, renders salinity tolerance a complex, multidimensional trait. A return to the genetic reservoirs of wild barley (Hordeum spontaneum) can serve as an effective strategy for broadening the genetic base of cultivated barley (Hordeum vulgare) and developing genotypes adapted to saline conditions. This study aimed to investigate the diversity arising from nested integration of the wild barley genome into a cultivated genetic background, to identify genetic resources and superior lines for salinity tolerance, and to elucidate the associated physiological mechanisms, through two studies. In the first study, the IWB-21 nested population (443 recombinant inbred lines derived from crosses between 21 wild barley genotypes and the cultivar Reyhan-03) was evaluated for agronomic and morphological traits across the entire population, and for physiological, ionic, and biochemical traits in 85 selected genotypes, under normal and saline conditions over two growing seasons. In the second study, 121 genotypes—including cultivars, gene bank accessions, selected recombinant lines, and wild barley—were examined under the same conditions over two growing seasons. Sowing in both studies was delayed (February 11), consistent with actual regional conditions in Isfahan province. Results from the first study showed that salinity significantly reduced most agronomic traits, including grain yield, biological yield, and harvest index, but the wide phenotypic variation in the IWB-21 population demonstrated its high potential for identifying adapted combinations. Transgressive segregation evidence indicated that recombination of wild and cultivated alleles can produce combinations superior to their parents; families 10, 13, and 15, along with lines RIL257, RIL283, and RIL405, were identified as superior genotypes. Physiologically, salinity was associated with reduced relative water content and potassium and increased proline, sodium, sodium-to-potassium ratio, and antioxidant enzyme activity, though superior lines showed adaptation more through a balance between yield and physiological response than through an intense defensive response. In the second study, recombinant lines with a mean grain yield of about 489 g/m² under saline conditions showed the lowest yield reduction (about 26%) and ranked most favorably on tolerance indices. RIL180, RIL287, RIL427, RIL247, and RIL281 were the top genotypes; RIL287, with only about a 3% yield reduction, was a notable example of yield stability. The composite significant index (CSI) showed a stronger relationship with actual yield under salinity than the yield stability index (YSI). Grain yield in both environments showed high heritability, and spike number per square meter was identified as a suitable indirect selection criterion under stress. Overall, the results showed that the genetic diversity of wild barley, particularly when transferred through nested populations and recombinant lines, holds considerable potential for improving yield and salinity tolerance, and that tolerance assessment should be based on a combination of yield, physiological traits, ionic balance, and multi-trait indices. The superior lines identified can be used in genetic studies, pre-breeding, and breeding programs.
