Abstract
Electrochemical glucose sensors estimate glucose concentration from the electric current generated by electrode reactions. Their response depends not only on glucose concentration but also on mass transport, reaction-region geometry, and electrode potential. This study investigates and compares the effects of these parameters on the amperometric response of a glucose sensor model. The reference Glucose Sensor model supplied with COM-SOL Multiphysics was reproduced using the finite element method. Simulations were then performed at glucose concentrations of 2, 5, 10, 15, and 20 mM, while varying the glucose diffusion coefficient, the simulation-domain height, and the working-electrode potential one factor at a time. The domain heights were 500, 1000, and 1500 micrometers, and the applied potentials were 0.35, 0.40, and 0.45 V. The model output was the mean current density at the working electrode. Performance was compared using the calibration slope (computational sensitivity) and the coefficient of determination (R²), which describes how closely the simulated points follow a fitted straight line. Over the tested parameter ranges, domain height had the greatest influence on the simulated response. Increasing the height from 1000 to 1500 micrometers increased the full-range calibration slope from approximately 2.15 to 6.11 (A/m²)/mM and R² from 0.78 to 0.97. Increasing the glucose diffusion coefficient increased current density, particularly at lower concentrations, but reduced the slope of the linear fit over the full concentration range. In contrast, changing the potential from 0.35 to 0.45 V changed the current density by less than 0.003%. Comparing two computational meshes at three domain heights produced differences below 0.4%, and additional numerical-setting checks preserved the main trends. To examine calibration linearity more closely, nine glucose concentrations between 2 and 10 mM were evaluated. R² was approximately 0.95 over this interval, increasing to about 0.997 for the five points between 2 and 6 mM. Applying the fitted straight-line equation to four additional concentrations without refitting yielded a maximum relative prediction deviation of about 1.9%. The findings show that a higher current response does not necessarily imply higher calibration sensitivity or better linearity. These conclusions are limited to the simulated model and conditions; no experi-mental or clinical validation was performed.
