Abstract Some of the members of Clavicipitaceae family are endophytic fungi which havemutualistic relationship with the plant family poaceae Their relationship benefits forboth host and fungus The endophytes spend all its life cycle on the aerial parts of planthosts and live intercellularly In the present investigation endophytic fungi were isolatedfrom seed and leaf sheath of the host Festuca arundinaceae Festuca avina Festucapratensis Bromus tomentellus Melica persica and Lolium prenne Morphologicalcharecters of the isolated fungi were checked on different culture media Most of theisolates were similar to the described genus of Acremonium of Albolanosa section and Neotyphodium from wild barley Sensitivity of the isolates to benomyle fungicide wastested and the isolates were catagorized into three groups of low medium and highlyresistant During performance of this examination it was found that media containingbenomyl enhance sporulation of the non sporulating isolates In antibiosis test it wasfound that most of isolates were effective against the phytopathogenic fungus Bipolarisaustralensis and only two isolates of FoGn and MpFn were effective against Pythiumaphanidermatum Genomic DNA was extracted using Chen et al and Liu et al methodsand three setsof primers ITSI ITS4 ISI IS3 and 111 112were used to detect and identifyendophytes Also RFLP PCR of ITS I and ITS2regions using Sau3A and Clo1 was carried 1out Direct sequencing of ITSI ITS2 and 5 8 S genes vIas performed Based onmoIphological and molecular characters the genusNeotyphodium on F arundinaceae F pratensis F avina B tomentellus M persica and L prenne and the N lolii on L prenneand N coenophialum on F arundinacea and Epichloefestucae on F ovina and closelyrelated speciesN bromicola on B tomentellus were identified It was also found that basedon only sequencing of ITSI ITS2 and 5 8 S gene regions it is not possible to have a clearview of the genome and for having more precise view of genome it is better to usesequencing of more regions and also use more restriction enzymes along withmoIphological characters 1 r Iii 1 Lf J 1 f I
