AbstractGenetic manipulation and selection of favorable somaclonal variant from callusculture are supplementary tools to traditional breeding for production of stress resistant in plants Response of twenty five cultivars of bread wheat Triticumaestivum L to immature embryo culture callus induction and regeneration fromcallus and in vitro salt tolerance with four different concentrations ofNaCI 0 5 10 15 gil was evaluated Callus induction rate and callus growth rate were usedto evaluate the responseof cultivars to immature embryo culture Also response ofcultivars to regeneration from callus was evaluated using the percentage and rateof plant regeneration Comparison of cultivars for salt tolerance at callus phaseaccomplished using callus growth rate relative growth and callus chlorosispercent cri eria Results of statistical analysis showed a highly significantdifferences among cultivars for response to callus induction Average percent ofcallus production ranged from 46 to 95 percent in Sholeh and Cross SorkhTokhm respectively There was a considerable genetic variation among cultivarsfor callus growth rate character which ranged from 0 31 in Cross Sorkh Tokhm to 0 21in Boomi Ilam and Ghods cultivars Evaluation ofcultivars response toregeneration from callus revealed a significant differences among cultivars Theaverage per centof regeneration ranged from 71 2 in Alvand to 13 7 in lnia Among the cultivars Navid was ranked the superior for regeneration from callusat the shortest time in comparison to other cultivars The results of the salttolerance experiment indicated that among the criteria were used for evaluatingcallus tolerance to salinity callus growth rate and percent of chlorosis are morereliable due to their highly significant negative correlation Sholeh cultivar hadthe highest callus rowth rate and the lowest callus chlorosis and was ranked asthe superior genotype for tolerating to salinity under in vitro condition Shahdas Roshan Ghods Mahdavi and Cross Sorkh Tokhm cultivars had highercallus growth rate and lower callus chlorosis and ranked as the second group ofsalt tolerant cultivars after Sholeh cultivar Zarrin cultivar had the lowest callusgrowth rate and the highest callus chlorosis and was ranked as the most sensitivegenotype for tolerating to salinity under in vitro condition There for it may bepossible to obtain cellular lines having salinity tolerance either due to superiorgene combinations or somaclonal variations by crossing the mentioned cultivarsand salt affected in vitro culture of resistant F 1hybrids t f or r I I
