Abstract In order to investigate the morphological biological and genetically relatedness of differentpopulations of Root Knot Nematode on pistachio during 2004 93 populations were collected frominfected roots and related soil These populations were obtained from different regions of Kermanprovince on different pistachio cultivars such as Fandoghi Akbari Ahmad aghaie kale ghoucheeand Badami Purification and multiplication of each population were done on two tomato cultivars Rutgers and Cormello Total DNA was extracted from eggs of all populations and two species ofRoot Knot Nematode Meloidogyne javanica and M incognita were identified using species specific primers The most cosmopolitan and widespread species was M javanica with 88 percentof whole collected populations Morphological and morphometerical characteristics of perinealpattern in M javanica populations revealed high variation among them Based on analysis ofmorphological qualitative characteristics 22 to 100 percent similarity was estimated among M javanica populations Analysis of morphometerical quantitative characteristics also confirmed thishigh level of variation between different populations of M javanica M incognita was limitedlydispersed in pistachio growing areas of Kerman province Perineal pattern of this species also leadto find a high level of variations Based on analysis of qualitative morphological characteristics 15 100 percent similarity was estimated among different M incognita populations The analysis ofmorphometerical characteristics also confirmed this variation among different M incognitapopulations Biological variation among these two species populations were also checked using hostdifferential test based on Saser and Carter method All M javanica populations could form galls ontobacco watermelon and tomato But the gall formation ability on pepper was variable and 48percent of M javanica populations could form gall on pepper This is the first report of Peppersuccessful parasitism by M javanica in Iran M incognita populations also showed differentinteractions with differential hosts IN4 population could have a successful parasitism and formgalls on pepper watermelon and tomato This population identified as Race 1 of M incognita Thisis the first report of Race 1 of M incognita on pistachio in Iran IN89 population couldn t form gallson cotton but was able to parasitize and form galls on other hosts and was identified as Race2 Other populations which were able to form galls on all differential hosts were identified as Race4 In order to investigate the genetically relatedness of M javanica and M incognita PCR RFLP andRAPD PCR methods were employed Among the primers used in this study the specific primerpairs OPARjav OPAFjav and Mjavf Mjavr amplified a 670 and 1600 bp fragments respectivelyfrom every M javanica populations Also a 500bp fragment was amplified using OPBFinc OPBRinc specific primer pairs in all M incognita populations RFLP profiles were obtained fromAluI TaqI Tru9I BamHI and PstI digests of the fragment from all populations in each species These profiles confirmed that populations in each species are identical Since only a limited regionof the genome is subjected to PCR RFLP analysis RAPD PCR that investigates the expanded areaof the genome was employed to find variation among the populations Rapid analysis wasperformed using 30 arbitrary decamer primers Cluster analysis based on RAPD profiles showedvariability between different populations in each species of M javanica and M incognita Therewere 91 and 86 percent similarity among different populations of M javanica and M incognitarespectively These results were in agreement with the results of similar studies on other hosts Based on the result of this study morphological biological and genetically variation amongpopulations of each species were proved but there was no complete relatedness between these threekinds of clustering which was similar to result of same studies Since morphological
