Abstract Pistachio known in Iran as Green Gold is a prominent product in terms of exportand foreign income due to its limited production in the world There are severalcultivars of pistachios with different names throughout different regions of Iran butthere is no information about their genetic relationships Addressing geneticrelationships amongst Iranian pistachios has been expected to have a marked impact onincreasing the productivity of this plant especially in terms of future production ofseedlings with favorable characteristics So far characterization of pistachios was basedon morphological isozyme and RAPD markers reported for a relatively small numberof cultivars Using other markers with higher reproducibility and polymorphism such asamplified fragment length polymorphism AFLP could improve our understandingabout genetic diversity of pistachios To this end we tested 13 AFLP primercombinations on 48 pistachio samples collected from Rafsanjan pistachio collection Several similarity matrices were constructed based on the Simple Matching Dice andJaccard coefficient Applying the unwieghted pair group method using arithmeticaverage UPGMA and complete linkage methods phylogenetic dendrograms weresubsequently constructed In addition to cluster analysis principle component analysiswas used to confirm the results of cluster analysis Data analysis was performed byNTSYS software ver 2 02 AFLP analysis produced 506 polymorphic bands with apolymorphism degree of 95 2 Based on cluster analysis pistachio cultivars weredivided into seven groups Most commercially cultivated pistachios were readily placedin group I and II Group III included two wild pistachios Baneh and Baneh Baghi Forremaining groups embraced 2 cultivated and 2 wild pistachios Interestingly 2 well known cultivated pistachios Lokesirizi and Vahedi were placed in two clusters distinctfrom most other cultivated pistachios Two wild pistachios Kasoor and P palaestina were found their places in two separated clusters with very low similarity withLokesirizi and Vahedi Analysis of morphological traits confirmed this AFLP basedclustering The clustering results separated cultivated pistachio according to twomorphological traits fruit shape and time of fruit ripening Group I included fruits withorbicular or elongated shapes whereas ovate shaped fruits fell in group II Early and lateripening fruits belonged to group I and II respectively It was hypothesized that theseprimers probably amplify regions of genome involving genes controlling these traits Nonetheless confirming the location of these genes and their relationship to the AFLPmarkers needs further research
