AbstractPathogenesis Related proteins are compounds is produced in response to pathogens orEnvironmental stress These proteins can be classified into seven teen group by aminoacid sequences serological differences and Enzymatic kinetic among these classes1 3 Glucanase and Chitinase are most studied be cause of 1 3 Glucanase byhydrolyzing1 3 Glucan bond and Chitinase by hydrolyzing 1 4 N acetyl amin bondcan directly hydrolize fungal cell wall and inhibit fungal growth The aim of thisresearch was Isolation over expression analysis of physiological activity of 1 3 glucanase from Medicago truncatula Until now numerous kind of 1 3 Glucanaseare Identified in various plants and thein fungicide effect were tested Watson et alreported 304 protein with peptide mass finger printing technique in medicagotruncatula among them a 1 3 Glucanase was registered with BF 650622 Id number But until now It s Biological activity had ot been studied In this case by Medicagosativa and Pisum sativum 1 3 Glucanase Gene alignment on Medicago truncatulaGenome three open reading frame with 50 similarity by having start and stop codonwas identified and isolated Then these fragments was cloned in PET21c vector andexpressed in E coli BL21 SDS PAGE technique reveal that proteins were produce asan Inclusion bodies We also test their fungicide effect after folding them to achievetheir natural structure on Alternaria alternaria and Fusarium graminearum Theresult were significant P 0 05 by SAS system analysis
