Abstract In recent years many attentions have been focused on determination of soilbiological properties The standard procedures for determination of soil biologicalproperties are well developed however to be routinely used in the soil laboratories there are some discrepancies Therefore many efforts have been spent to developalternative simple and rapid procedures providing similar accuracy and precision Thenewly developed procedures are usually less time consuming labor intensive andprovide less exposure to toxic chemicals Microwave irradiation has been used as analternate energy source for many experimental procedures This source of energy canfacilitate some soil biological assays The objectives of this study were to identifywhether the microwave irradiation can be used as an alternative to the chloroformfumigation in soil microbial biomass determination the autoclave assisted digestionof soluble organic N in soil extracts and the prolonged incubations usually perform inN mineralization potential For these purposes soil samples were collected fromdifferent climatic regions of Iran General soil properties including pH ECe organic C calcium carbonate equivalent and soil textures were determined Soil microbial biomassN was measured by either chloroform fumigation extraction CFE or microwaveirradiation extraction MWE techniques The soluble organic N in the soil extractswere measured by both alkaline persulfate and microwave assisted oxidation Potentially mineralizable N was also measured either by anaerobic incubation or by amicrowave based technique Results indicated that at w w water content jg soil can eradicate microbial activities without disturbing soil humic material Following optimization of the microwave irradiation a significant r P correlation was observed between the values of soil microbial biomass N measured byCFE and MWE techniques suggesting that both techniques have determined similarpools of soil N In order to determine soluble organic N in the soil extracts we observedthat reagent sample ratio along with j g irradiation in two equal j g bursts can provide highly significant correlation r P and identicalresults compared to the standard alkaline persulfate oxidation procedure In contrast microwave irradiation did not perform satisfactorily in estimating potentiallymineralizable N since the correlation between the anaerobic incubation and microwave based technique was not significant r Overall our results clearly demonstratedthat microwave irradiation can be used as a simple rapid and non toxic procedure forhazardous time consuming chloroform fumigation Similarly it can be satisfactorilyused routinely for precise determination of soluble organic N Microwave irradiation asan alternate source of energy for autoclave accelerated oxidation of different N forms inthe soil extracts to nitrate However we could not recommend microwave irradiation asan alternate procedure for prolonged incubations which are traditionally used in Nmineralization determination Finding a proper microwave assisted technique for Nmineralization warrant further researches
