Study of Response of Melon Cultivars to Tissue Culture in View Point of Organogenesis and Embryogenesis Reihane Moradmand Riehane Moradmand@yahoo com 16 3 2009 Department of Agronomy and Plant Breeding Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language FarsiSupervisors A Arzani a arzani@cc iut ac ir and G Saeidi gsaeidi@cc iut ac ir AbstractMelon Cucumis melo L is an important horticultural crop worldwide Melons are cross pollinated witheconomic consequence that their cultivation are common in many parts of Iran This vegetable crop isuseful foodstuff in diet due to it s glucose vitamin mineral and antioxidant Due to heterozygosis natureof melon it s conventional breeding restricted and application of biotechnology techniques such as tissueculture is necessary Cultivation of exotic variety and huge genetic erosion in domestic germplasm inrecent years the danger of distinction of native germplasm is inevitable in melon Therefore use ofmicropropagation technique for proliferation of melon is essential Organogenesis and somaticembryogenesis are used for plant regeneration from tissues in this crop This research assessed desirableconditions for organogenesis using cotyledon leaf and root explants in three genotype of melon includingIsfahan melon Contaloup and Gargar Explants cultured in Murashig and Skoog MS medium and thereaction of explants was evaluated Root explants was not able to regenerate Cotyledon explantsindicated good potential in MS supplemented with BA and produced high shoot induction in MS with1mg lit BA and 0 5mg lit NAA for induction root High shoot induction 2 5 shoot in each explants observed in MS with 1mg lit BA in Isfahan melon genotype Isfahan melon genotype possessed superiorreaction to organogenesis The highest rate of shoots and number of leave was observed in cotyledonexplant Regeneration of shoot from leaf explants was low Inact embryonal axis and cotyledon explantswere culture on MS medium with BA in combination 2 4 D at different concentration 2 4 D is necessaryfor callus induction One mg lit 2 4 D with 0 5 mg lit BA produced the highest induction embryo and 2 4 D concentration influenced the rate of callus growth Steps of embryo growth included globular heartshape torpedo and cotyledonary and abnormal forms of embryo and meristemis region growth observedin this research indicated that BA necessary for shoot induction and 0 5 1 mg lit BA was optimal toinduce somatic embryo 2 4 D combined with BA were effective on callus induction In conclusion genotype explants and plant growth regulators were effective in response to tissue culture in melon Key Word Cucumis melo L micropropagation Organogenesis somaticembryogenesis regeneration cotyledon
