Evaluation of Intrapopulation Microsatellite Genetic Diversity among Iranian Pistacia Genotypes Haleh Salimi haleh salimi @gmail com 9 3 2009 Department of Agricultural Biotechnology Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language Farsi1 M Bahar Assoc Prof Supervisor mbahar@cc iut ac irAbstractThe genus Pistacia is a dioecious and wind pollinated member of the Anacardiaceaefamily that consists of 11 species Among these species Pistacia vera has edible nutsand commercial importance The other species usually grow wild and are used asrootstock for P vera Principally the most typical wild Pistacia species in Iran areP atlantica P khinjuk and P vera L var sarakhs It is suggested that P vera is themost ancestral species of Pistacia and the other species are probably its derivatives Pistachio P vera L cultivation and production has a long history in Iran and thecountry is well known internationally for this enterprise Production and exportationof this product in spite of the presence of international competitors is comparativelybeneficial and plays an important role in attracting foreign currency and attainment ofeconomic trading status Natural forests of wild Pistachio trees are still seen indifferent areas in North East of Iran and Afghanistan They are valuable geneticresources and their genes can broaden the Pistachio gene pool and make the breedingprograms more efficient Moreover Pitachio ability to tolerate salinity and droughtconditions has made it a suitable crop for many regions of Iran The multiple namesfor commercially grown Pistacia cultivars utilized in different cultivation regions ofIran have somewhat created confusion for precise genotyping Patterns of geneticdiversity in Pistacia have been studied using a variety of morphological physiological and biochemical methods Morphological characteristics vary amongspecies However for many of the species these distinctions are more difficult toevaluate There is almost no cytogenetic information available on the various Pistaciaspecies Different researchers have conducted isozyme studies with the aim ofdistinguishing varieties of P vera but insufficient isozyme polymorphism amongclosely related cultivars has limited the usefulness of isozymes for analysis of geneticdiversity and relatedness The problem of insufficient polymorphism can be mitigatedby using DNA based markers In this study we attempted to 1 Demonstrate thecharacteristics of the trinucleotide AAG 8 microsatellites in order to design SSRprimers for the Pistacia genome 2 Use microsatellite markers to identify differentPistacia species 3 Characterize a germplasm with a wide range of pistachiogenotypes originated from Iran Syria Turkey and United States using SSR and ISSRmarkers 4 Investigate the phylogenetic relationship between genotypes collectedfrom different countries In this study plant material from commercially grownPistachio cultivars was collected from four different countries including Iran Syria Turkey and United States of America Primiraly An enriched library for AAG 8repeat motifs was constructed from genomic DNA of Ohadi cultivar and one ofP khinjuk accessions following the procedure of Hamilton et al 1999 with minormodifications The fragments containing microsatellite repeats identified and then
