Cultivation and growth of Scenedesmus quadricauda at differentconcentrations of nitrogen for application in aquaculture and wastewater treatment Safiolla Heidari E mail safiolla heidari@yahoo com September 2009 Department of Natural Resources Isfahan University of Technology Isfahan 84156 83111 IranDegree M sc Language FarsiO Farhadian Assist Prof Supervisor omfarhad@cc iut ac irAbstractNitrate and ammonium as nitrogen source were used by microalgae in waterbodies They play important roleeither in removing of nitrogenous compounds or in production of useful algal biomass The species with hightolerance for nitrogen compounds are useful in order to treating nitrogen rich wastewaters These benefitsmake microalgae as practical organisms in aquaculture activities Therefore introduction of the indigenousmicroalgae named Scenedesmus quadricauda which has suitable growth and reproduction on nitrogen richwastewaters was selected for this research In this research firstly S quadricauda was cultured underdifferent nitrate and ammonium levels 2 9 to 150 mM as nutrient and then in another experiment efficiency of this algae was examined in order to remove ammonia and nitrite compounds from fish farmeffluent Results showed that dry biomass of S quadricauda for 2 9 25 50 75 100 and 150 mM of NaNO3were 0 89 0 86 0 48 0 48 0 26 and 0 22 mg ml respectively while for the same concentration of NH4Cl dry biomass were 0 89 0 76 0 53 0 38 0 38 and 0 31 mg ml respectively In the other hand specificgrowth rate SGR for the same concentration of NaNO3 were 0 065 0 063 0 015 0 015 0 045 and 0 065day 1 respectively while in the same concentration of NH4Cl SGR were 0 065 0 053 0 023 0 003 0 003and 0 023 day 1 respectively Dry biomass of S quadricauda for 2 9 5 10 15 20 and 25 mM of NaNO3were 0 54 0 68 0 75 0 78 0 68 and 0 50 mg ml respectively while for the same concentration of NH4Cl dry biomass were 0 54 0 65 0 50 0 39 0 38 and 0 37 mg ml respectively SGR for the similarconcentration of NaNO3 were 0 192 0 230 0 240 0 273 0 260 and 0 185 day 1 respectively while for thesame concentration of NH4Cl SGR were 0 192 0 143 0 143 0 170 0 083 and 0 059 day 1 respectively Therefore the best range for growth of this algae is between 2 9 to 25 mM of NaNO3 and NH4Cl while theoptimum concentration of NaNO3 and NH4Cl was obtained 15 mM In this concentration S quadricauda hadmaximum SGR 0 27day 1 and minimum population doubling time DT 2 54 day Our findings aboutculture of S quadricauda on different wastewater treatments indicated that better growth performanceobtained with diluted fish farm effluent ammonia 0 11mg L nitrite 0 62 mg L In these conditions maximum cell number 9 106cell ml maximum chlorophyll a 2 90 mg L and better ammonia 89 57 and nitrite 89 53 removing were found Results of this research indicated that S quadricauda assimilatenitrogenous compounds in natural waters and produce efficient algal biomass in culture tanks This algaecould be used as suitable live food in zooplankton culture in field and laboratory conditions In addition thisspecies is a suitable candidature to design of bio filter systems which can remove nitrogenous compoundsfrom wastewaters and effluents resulted from anthropogenic and agricultural activities Key wordScenedesmus quadricauda BBM nitrate ammonium specific growth rate chlorophyll a wastewater treatment
