Combination of hot water and surfactant assisted dispersive liquid liquid microextraction for determination of sulfonamides in chicken meat and water samples by high performance liquid chromatography Behnaz Harandizadeh Behnaz harandizadeh@yahoo com April 5 2010 Department of Chemistry Isfahan University of Technologhy Isfahan 84156 83111 Iran Degree M SC Language Farsi Supervisor Assist Prof Mohammad Saraji saraji@cc iut ac ir Abstract In this work hot water extraction and dispersive liquid liquid microextraction coupled with high performance liquid chromatography diode array detector HPLC DAD were applied for the extraction and determination of sulfonamides sulfacetamide sulfadiazine sulfamethoxazole and sulfaquinoxaline in chicken meat and water samples For the microextraction extraction solvent chloroform contains 5 surfactant methyl trialkyle ammonium chloride was injected in to a 10 mL glass tube with conical bottom containing aqueous solution of sulfonamides by a 50 L syringe After centrifugation the sedimented phase was complectely transferred to another tube with conical bottom using a 25 L HPLC syringe 10 L mixture of methanol water 30 70 was added in to the sedimented phase and finaly 20 L of the solution was injected into the HPLC Extraction of these compounds from the meat was performed with hot water at 100 oC In order to obtain optimum conditions of the microextraction several parameters affecting on the microextraction such as extraction solvent type percentage of disperser solvent in extraction solvent pH sedimented phase volume salt addition and time of the extraction were studied Parameters affecting on the extraction of these compounds from the meat contaning temperature time pH and volume of water were studied Under optimum conditions for microextraction extracting solvent chloroform amount of surfactant in chloroform 5 v v sedimented phase volume 10 L pH 7 without salt addition Enrichment factors and limit of detections LODs were ranged 27 9 56 7 and 0 01 0 2 g L respectively The relative standard deviations R S D s for analysis of 20 g L of sulfonamides in water samples were in the range of 3 6 5 3 n 5 Optimum condition for extraction of these compounds from the meat were achieved as 10 mL buffer pH 4 at 100 oC for 10 minutes The limit of detections LODs for chicken meat were in the range of 25 8 40 3 g kg As a result the proposed method was used for analysis of rael samples such as zayandeh rood Esfahan river water fish pool water native chicken meat and chicken meat from Homa and Esfahan company Keywords Dispersive liquid liquid microextraction Diode array detector Sulfonamide Hot water Chicken Meat
