Transition and Expression of Albugo candida Pathogenicity Candidate Molecules in some Model Plant Javad Mirzabeigi j mirzabeigi@ag iut ac ir Date of submission June 15 2010 Department of Agricultural Biotechnology Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Language Farsi 1 B Sharif nabi Assoc Prof Supervisor Sharifna@cc iut ac ir 2 M H Borhan Assist Prof Supervisor Mhborhan@yahoo com Abstract The fungal like organism Albugo candida pers Roussel from family Albuginaceae order Albuginales class peronospromycetes from oomyceta is the casual agent of white rust of canola This branch of fungi produce two types of pathogenic factors for pathogenicity in host plant some groups act as intercellular elements and some as intracellular ones To study the pathogenic factors it is necessary to express the genes encoding these proteins in plant Due to the lack of information regarding to pathogenic factors and effective genes in disease caused pathogenicity conducting a research regarding the matter seemed to be essential In this study the number of 50 constructs containing candidate genes have been used for transient expression in Arabidopsis thaliana Nicotiana benthamiana and Brassica napus which expected to produce effective molecules of A candida These samples had been isolated from cDNA library of this fungal like by Dr Borhan from Saskatoon university After transformation by agroinfilteration in Arabidopsis and tobacco non of the samples caused morphologic signs in these plant In canola both of the constructs showed the disease signs chlorosis Then eight construct were sequenced Two sequences which showed the disease signs were compared with other pathogenic proteins containing Nep1 domain by Blastx and Blastp which pointed the adaptation By signalp software the existences of marker peptide were verified in these two sequences that causes the secretory property in protein Therefore the sequences which were adapted with two pathogenic sequences by doing
