Chemiluminescence Trace Determination of Glutathione and Isoproterenol Using Luminol Diperiodatoargentate III System and Study of the Catalytic Effect of Beta Cyclodexterin CD MnFe2O4 MNPs on the Speed of Determination Fariba Haghighatnia February 2011 Department of Chemistry Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Lanquage FarsiSuperviser Bahzad RezaeiEmail E mail rezaei@cc iut ac irAbstract The level of glutathione GSH is important in blood and urine as biomarker in a number of clinicaldiagnoses It is the major endogenous antioxidant produced by the cells participating directly in the neutralization offree radicals and reactive oxygen compounds Through direct conjugation it detoxifies many xenobiotics Thisincludes heavy metals such as mercury lead and arsenic Isoperterenol ISP is a drug that plays major role in thetreatments of bronchi constriction in patients with asthma and may be used in emergency patients with heart block Therefore determination of GSH and ISP is important In recent years enormously sensitive analytical techniquesbased on chemiluminescence CL systems have received noticeable attention Simplicity of detection low detectionlimit wide linear dynamic ranges and short analysis times and relatively low cost of instrumentation are some ofthe characteristics that make this methods appealing In this work CL method has been used for the determination ofGSH and ISP It is based on the enhanced CL of the reaction of luminol and diperiodatoargentate III DPA K Ag H3IO6 2 in an alkaline solution At first the effects of chemical and instrumental variables on the CL intensitywere studied In chapter 1 in GSH determination the catalytic effect of inclusion complex of beta cyclodexterin CD MnFe2O4 MNPs magnetic nanoparticles in different medium on the CL response of luminol DPA systemwas investigated The MnFe2O4 MNP S were prepared at physics laboratory of Isfahan University of Technology Cyclodextrins have been used extensively in luminescence techniques due to the fact that some analytes cause theluminescence intensity to change by making complex with cyclodextrins The CD formed an inclusion complexwith MnFe2O4 MNPs via absorption on the surface of MnFe2O 4 MNPs which could stabilize them In recent years there have been paid considerable attentions to the metallic and magnetic nanoparticles MNPs because of theircatalytic effect super paramagnetic behavior their large ratio of surface to volume and low toxicity Using thiscatalyst dramatically make the CL intensity increase 9 times and response time the time that peak was reached itsmaximum level decrease about 10 times Under the optimum conditions dynamic ranges were obtained from therange of 5 0 10 8 to 4 0 10 6 mol L of GSH The detection limit of the method is 1 5 10 8 mol L 3S b m and therelative standard deviation for 9 replicates measurement of 8 0 10 7 mol L GSH was 2 8 The proposed method iscompared with the Ellman reference method for the determination of GSH It was successfully applied for the rapidand sensitive determination of GSH in human blood samples In chapter 2 the linear correlation between the CLemission intensities and ISP concentrations was investigated Under the optimum conditions the CL intensity vs ISP concentration was linear in the range of 1 0 10 8 to 9 0 10 7 mol L The detection limit was 4 7 10 9 mol Lfor ISP and the relative standard deviation for n 11 replicates measurement of 2 0 10 7 mol L was 2 7 Theproposed system was successfully applied to determine of ISP in human serum and pharmaceutical The reactionmechanism for these CL reactions are proposed and discussed Keywords Chemiluminescence inclusion complexes CD MnFe2O4 MNPs magneticnanoparticles Glutathione Human blood Luminol Diperiodatoargentate III Isoproterenol Human plasma Pharmaceutical
