Determination of Genetic Diversity in Pomegradnate Punica granatum Using Chloroplast Microsatellites Mohsen Nourouzi Mohsen88nourouzi@yahoo com Date of submission August 28 2011 Department of Agricultural Biotechnology Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Language Farsi W M Talebi Supervisor mtalebi@cc iut ac ir d W 2 Supervisor sayedt@cc iut ac irAbstractPomegranate Punica granatum L is one of the oldest fruits known to mankind and is one of the mostimportant endemic horticultural plants in Iran According to historical evidences it seems that Iran hasbeen the origin of pomegranate and it has spread from this region to the other parts of the world Longhistoric cultivation of pomegranate in Iran has led to synonymies and homonymies among genotypescultivated in different regions A precise method for determination and discrimination of the genotypes isrequested for breeding and commercialization of Iranian pomegranate cultivars and it can be achievedusing molecular markers and morphology characters With the development of modern biotechnology molecular markers are used more than morphology character because they are not affected byenvironment Molecular studies of the pomegranate have been restricted to examinations of nucleargenomic to investigate the population dynamics of these economically important cultivars Among all ofthese molecular markers in nuclear genome microsatellite markers showed higher efficiency than othersto separate the pomegranate genotypes The SSR markers derived from chloroplast genome generallyshow simple uniparental modes of inheritance haploid nature and high copy number which make thembetter than genomic microsatellite Therefore to characterize the genetic diversity and populationstructures of pomegranate in Iran cpSSRs were studied on 52 cultivars from different geographicalregions to reveal the cpDNA variation among and within populations In the present research theinvestigation of the genetic diversity of pomegranate was done based on cpSSRs markers on 52 Iraniangenotypes of pomegranate among which 16 cpSSR primer pairs amplified 21 loci and 16 loci 80 wereshown to be polymorphic The average number of alleles for each cpSSR locus was 2 44 The averagevalues of PIC and Nei s gene diversity He for all loci were 0 24 and 0 37 respectively The copheneticcorrelation coefficient based on Dice s similarity coefficients by UPGMA algorithm was accounted high r 0 88 indicating that the dendrogram was a good representation of the similarity matrix Clusteranalysis was able to cluster 52 genotypes based on cultivation somedeal as cultivated and wild genotypesalmost classified into separate groups The total ornamental genotypes stand into a subcluster participantwith other genotypes Pomegranate genotypes were classified based on cultivation and geographicalregions to analyze the genetic diversity within and among groups and performed the analysis of molecularvariance AMOVA which results showed a significant difference among groups The result ofPOPGENE analysis based on cultivation demonstrated a high degree of similarity among groups Maximum similarity was observed between wild and cultivated groups This research indicates that thelevel of polymorphism among pomegranate genotypes is appreciably high in Iran and also the results ofthis study suggested that cpSSR marker offers a powerful means to analyze the genetic diversity withinand among populations Keywords Punica granatum cpSSR Genetic variation AMOVA
