An investigation on producing of sericin nano powder through electrospraying technique Najmeh Hazeri n hazeri @tx iut ac ir Date of Submission 2011 10 11 Department of Textile Engineering Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language PersionSupervisors Dr Hossein Tavanai tavanai@cc iut ac ir Dr Alireza Moradi moradi a@cc iut ac ir AbstractSilk is a natural protein that is mainly made of sericin and fibroin proteins Sericin constitutes25 30 of silk protein and it envelops the fibroin fiber with successive sticky layers that help inthe formation of a cocoon Most of the sericin must be removed during raw silk production Atpresent sericin is mostly discarded in silk processing wastewater If this sericin protein isrecovered and recycled it can represent a significant economic and social benefit The sericinprotein resists oxidation is antibacterial UV resistant and absorbs and releases moisture easily Sericin protein can be cross linked copolymerized and blended with other macromolecularmaterials especially artificial polymers to produce materials with improved properties sericin inthe powder form has found a wide range of applications such as food and cosmetics additive aswell as drug delivery Generally speaking powder can be produced through a number of techniquessuch as Precipitation of particles with a coagulant agent in sericin solution and spray drying ofsericin solution Electrospraying is a method for production solid nano particles The literaturereview showed no report on the production of sericin nano powder through electrosprayingtechnique So in the present study the production of sericin nano powder through electrosprayingwas persued To produce sericin nano powder through electrospraying at first sericin wasrecovered from bombyx mori cocoons at high temperature and high pressure HTHP It is believedthat high temperature and high pressure method yields the highest protein recovery amongst all themethods used to recover the sericin from the cocoon It is worth mentioning that enzyme process isonly used in the degumming process The main aim of degumming process is to obtain the silkfilaments for mainly the textile industry applications and the sericin itself is usually discared aswaste water The solution obtained was centrifuged and the sericin solution was dryed by means offreeze dryer and the sericin in the solid form which is also called sponge was obtaind To preparethe electoapraying solution The sericin sponge was dissolved in dimethylsolfoxide DMSO andelectrospraying was carried out for a set of conditions including voltage needle collector concentration and feed rate as the important factors affecting the electrosprayed sericin nanoparticle size The levels for each of these factors were chosen by trial and error The result of thisstudy showed that electrospraying is capable of producing nano powders with average particlessize less than 80 nm as well as having uniform spherical shape Also it was ascertained that lowerconcentrations lower feed rates longer needle collector distances lead to a decrease in the averageparticle size of sericin nano powder Voltage decreased the particle size up to 20 kV and with highervoltages the average size increased FT IR and XRD studies showed that the structure ofelectrosprayed sericin is similar to spone sericin but with a lower crystallinity index The resultalso showed that regain humidity of sericin nano powder is more than spone sericin The ultraviolet UV studies showed that the electrosprayed sericin nano prticles had a higher UVabsorbance than the sericin sponge Key words sericin electrospraing nano powder particle size
