Biological Removal of Polycyclic Aromatic Hydrocarbons PAHs from Contaminated Water in Fixed Bed Fungal Biofilm Reactor Mahsa Baniasadi Baniasadi mah@gmail com Data of submission 11 9 2011 Department of Chemical Engineering Isfahan University of Technology Isfahan 84156 8311 IranDegree M Sc Language FarsiSupervisor Hamid Zilouei hzilouei@cc iut ac ir Seyed Abbas Shojaosadati shoja sa@modares ac irAbstractPolycyclic Aromatic Hydrocarbons PAHs comprise a large and heterogeneous group of organic contaminantsthat are formed and emitted as a result of incomplete combustion of organic material or derived from industrialactivities thus soils from gasworks sites and carbochemical plants as well as refineries and filling stations areoften contaminated with PAHs With their carcinogenic and mutagenic properties PAHs are recognized as apriority pollutant by environmental agencies There is a toxicological concern about the presence of PAHs in theenvironment Application of biological treatment system using white rot fungi became an interesting alternativefor treatment of water or wastewater contaminated with persistent organic pollutant such as polycyclic aromatichydrocarbon The lignin degrading white rot fungi Phanerochaete Chrysosporium has the ability to degrade awide variety of organopollutants due to its non specific extracellular enzymes It has been shown that polycyclicaromatic hydrocarbons PAHs can be oxidize by P chrysosporium The best characterized white rot fungalextracellular enzymes are lignin peroxidase LiP Mangenese peroxidase MnP and laccase Immobilization ofmicroorganism in biofilm is appropriate for use in environmental biotechnological processes In environmentalbioprocess unlike other industrial bioprocesses large volumes of dilute aqueous solution have to be treated andthe processes need to be operated at high biomass without the need to separate the biomass and treated effluent Packed bed bioreactor using white rot fungi Phanerochaete chrysosporium was evaluated for degradation ofPAHs in synthetic polluted wastewater By means of their ligninolytic enzymes LiP MnP and laccase whiterot fungi have significant potential to metabolize organic pollutants such as PAHs In this thesis evaluation andoptimization of pollutant removal process conducted in different feed flow rates as well as initial PAHsconcentration in feed Pollutants concentration pyren and phenanthrene and flow rates varied from 50 to 100ppm and 0 14 to 0 55 ml min respectively and application of packed bed bioreactor for bioremediation wasconfirmed The result shows removal efficiency of up to 98 and enzyme activity of 57 and 426 Ul 1 for MnPand LiP respectively in optimum condition Variable such as MnP and LiP activity and pyren and phenanthreneconcentration was modeled In constant flow rate enzymes activity decreased by increasing pollutant initialconcentration In constant pollutant concentration by decreasing flow rate residual pollutant concentrationdecrease which means increase of removal rate Residual pollutant concentration measured in optimumcondition over time The system reaches steady state condition after 9 hours System operated properly for 7days After 7 days the reactor was clogged by large amount of biomass and needed to be shut down Themeasurement of oxygen consumption in reactor confirmed that degradation occurred biologically Keywords polycyclic aromatic hydrocarbons Phanerochaete chrysosporium packed bedreactor manganese peroxidase lignin peroxidase
