83 Effect of Glycinebetaine on Germination and Growth of Sweet Corn Under Salinity Stress Shahrbanoo Garmsiri s garmsiri@iut ac ir Date of Submission Jun 02 2013 Department of Horticultural Science Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language FarsiSupervisors A A Ramin aa ramin@cc iut ac ir F Amini F Amini@araku ac irAbstract Salinity is one of the most common abiotic stress considerabley reduced plant growth and yield of more plant species Decrease in torger potential caused by salinity is the main reason of reducing in plant growth under saline conditions One of the strategies that plants get ahead to adaptation with adverse environmental conditions such as salinity is accumulation of low molecular weight compounds that named compatible solutions Glycine Betaine is one of the most important osmolytes that play major role in osmoregulation in plants However in many crop plants the natural synthesis accumulation of Glycine Betaine is lower than the requisite level to ameliorate the adverse effects of various environmental osmotic stresses whit this reason an number of researchers believe that a method of enhancement of endogenous levele of plants Glycine Betaine is exogenous use of this material Therefore in order to investigate effect of this substance on the alleviation of salinization effect on sweet corn germination seedling emergence and vegetative stages three experiments was carried out in Isfahan University of Technology research greenhouse in year 2012 Experiments was carried out in completely randomized design CRD into factorial with two factors Glycine Betaine and salinity In germination and seedling emergence experiments Glycine Betaine in five levels 0 5 10 20 and 40 mM and salinity in tow levels 0 and 60 mM and with 5 and 3 replications in germination and seedling emergence stages respectively Seeds were soaked before planting for 24 hours in a solution in mentioned concentration of Glycine Betaine Vegetative stsge experiment was carried out in poty mode with tree Glycine Betaine levels 0 25 and 50 mM and three salinity levels 0 30 and 60 mM with 4 replications Glycine Betaine treatment in foliar spraying method was done in three stages with an approximately interval of 10 days The results showed that salinity treatment was ineffective on germination percentage but diminish germination rate and treatment seeds with Glycine Betaine had not a positive effect on this parameter However the results of seedling stage showed that the salinity negatively affected most measured parameter but seed treatment with Glycine Betaine could reduce salt damage truly In this experiment the concentration of 10 mM Glycine Betainen in absence stress condition and 20 mM in salt stress conditions had more alleviation effect Compared with other leveles Results of vegetative phase also showed that salinity led to adverse effectes on growth and growth parameters So that the plant height shoot and root dry weight relative chlorophyll chlorophyll a chlorophyll b and total chlorophyll leaf proline concentration concentration of sodium and potassium ions in leaves and roots ion leakage leaf anthocyanin and ascorbate peroxidase and peroxidase enzymes were affected by salinity But spraying the plants with Glycine Betaine improves the damage was caused by salt on most parameters so that the plants were sprayed with this material significantly have more height fresh shoot weight dry root weight chlorophyll a chlorophyll b total chlorophyll anthocyanins root s potassium ascorbate peroxidase and peroxidase enzymes and less proline and shoot s and root s sodium than non treated plants Overall according to obtained results It can say that in salinity conditions spraying with concentration of 50 mM Glycine Betaine enhanced growth and tolerance to salinity in sweet corn plants Keywords Sweet corn Salinity stress Glycine Betaine Germination stage Seedling stage Vegetative stsge
