Isolation of VvDREB gene from grapevine and its overexpression in model plant Tobacco to increase osmotic stresses tolerance Elham Mahvelati elhammahvelaty@gmail com Date 2013 1 17 Department of Agricaltural Biotechnology Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language PersianBadraldin Ebrahim Sayed Tabatabaei Supervisor sayedt@cc iut ac irAbstractSome common environmental stresses such as drought salinity and cold adverselyaffect plants to perform their complete genetic potential and have constraint effects oncrop growth and yield Plants may tolerate and adapt to these stressors by mechanismsat physiological biochemical and molecular levels Molecular studies have shown thatmany genes can be induced by stressors through stress signaling and gene expression The expressed genes based on their characteristics and the products can be categorizedinto two groups including those encode functional proteins and the genes that theirproducts are regarded as the most important components for deciphering signaltransduction of stresses and expression controls of stress tolerant genes Transcriptionfactors are critical regulators of gene expression system that play central roles inregulating downstream genes contributing to stress tolerance and the regulatory role ofDREB transcription factors have been extensively studied DREB transcription factorsare one of the four independent regulation systems in plants which involves in abioticstress signaling pathway and activation of stress responsive genes The objectives ofpresent study were to identify isolate and investigate the VvDREB and orthologousSbDERB2A genes from grapevine Vitis vinifera L Askari and the constitutiveexpression of VvDREB gene under salt stress conditions in Tobacco model plants Grapevine vitroplants were subjected to salt stress 150 mM NaCl for 24 hours andtotal RNA was extracted Specific primers were designed based on grapevine s droughtEST and SbDREB2A gene homologue in halophilic plant Salicornia brachiata VvDREB and VvsDREB DREB like 2C genes were isolated from grapevine Vitisvinifera L Askari Bioinformatics results indicated that VvDREB gene has 85 similarity with DREB2 genes from soybean family and VvsDREB gene has 80 similarity with SbDREB2A gene SbDREB2A and VvsDREB proteins motif studies inELM database showed that there are no five motifs BRCT MAPK PKB NES NLS in VvsDREB protein A cassette consisting of grapevine VvDREB cDNA driven byCMV35S promoter was cloned into pBI121 binary vector and transferred intoAgrobacterium tumefaciens strain LBA4404 Leaf disks from vitroplants were co cultivated with A tumefaciens and transformants under salt stress and water deficitconditions were selected The presence and expression of the gene in transformantswere confirmed by molecular methods PCR and RT PCR techniques respectively Increasing tolerance to salinity and drought were observed in transformed plantscompared with the control plants Overall it could be concluded that the VvDREB genebelongs to DREB2 family in plants Keywords Vitis vinifera L DREB Salt stress Water deficit Salicornia brachia
