Determination of tramadol in biological samples by combining headspace solid phase microextraction and ion mobility spectrometry Afshin moradi Afshinmoradi65@rocketmail com December 10 2012 Department of Chemistry Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Lanquage FarsiSuperviser Mohammad Taghi Jafari Email jafari@cc iut ac irMohammad Saraji Email saraji@cc iut ac irAbstract In this thesis sol gel process is used for manufacture of polydimethylsiloxane adsorbenton steel wire After that tramadol drug was extrated by the solid phase microextraction andanalyzed by Ion mobility spectrometry An injection chamber was constructed forconnection of SPME technique to ion mobility spectrometry In this design sample input gas inlet and element location is on one piece In this project the dead volume of injectionchamber is low so that the sample leaves the injection chamber and quickly enterd to theion mobility spectrometer Therefore the peak broading can be minimized Extractionparameters such as salt concentration temperature equilibrium and extraction time werestudied The best extraction efficiency was observe at saturated salt concentration 15 mMof NaOH Temperature was 97 C equilibrium time was 2 min and the extraction time was10 min Calibration curves were plotted in water urine and plasma sampels For water andurine samples linear daynamic range was obtained from 2 100 g L and r2 was 0 9982 and0 9976 for water and urine sampels respectively Linear daynamic range was obtainedfrom 20 to 700 g L and r2 equal 0 9980 for plasma sample The limit of detection wasobtained 1 g L and 9 g L in analisis of urine and plasma samples respectively Relativestandard deviation in urine and plasma samples was 8 and 10 respectively For theability of the method in analysis of real samples urine and plasma samples of volunteeranalyzed by the above method after toking tramadol Key Words Solid phase microextraction Sol gel Tramadol Injection chamber Ion mobilityspectrometry
