129Assessment of Essential Oil Composition and Genetic Diversity of Iranian Fennel Foeniculum vulgare Mill Using SRAP and Morphological Markers Hossein maghsoudi kelardashti Hossein maghsoudi65@yahoo com Submitted on 2013 Department of Agronomy and Plant Breeding Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Language Farsi M Rahimmalek Assist Prof supervisor mrahimmalek@cc iut ac ir Abstract Medicinal plants play important role in communities hygienic and health both in terms of disease cure and preventing Medicinal plants in Iran serve as valuable genetic resource Among these plants Fennel Foeniculum vulgare Mill belonged to Apiaceae family has received great deal of attention The previous studies in Iran and worldwide have mentioned enormous properties for fennel Various climate conditions in Iran fennel has high genetic diversity So managing of genetic resource knowledge of intra and interspecific diversity will help getting information about evolutionary processes and genetic erosion about this valuable species The most principle goal to conduct this research is to evaluate genetic variation of fennel endemic accession using molecular marker of SRAP and also classification of accessions in respect to essential oil percent major compounds using gas chromatography mass spectrometry In addition self and cross pollination status and morphological traits were also investigated in fennel This study was carried out in research farm of Isfahan university of technology where located at Lavark region in completely randomized block design in thrice replicates in agronomic years of 2010 2012 Twelve markers combinations of SRAP produced 192 polymorphic bands in which of them were polymorphism percent and PIC were respectively 88 41 and 0 36 Analysis of molecular data classified 55 fennel genotypes into five classes based on geographical position and climate Adjusted principle coordinate analysis PCoA also confirmed cluster analysis results The main composition and substance in fennel leaves was Trans anetole so that the highest amount of this compound was in Shiraz accession 56 61 and the lowest one was observed in Shirvan sample 41 19 while the highest and the lowest essential oil percent were recorded in Tabriz 2 03 and Isfahan 0 65 respectively Sample classification based on essential oil and major compounds was in line with molecular classification Based on results obtined GC MS analysis limonene fen chon Trans anetole were the most frequent substances in tested essential oil and based on this twelve fennel accessions were grouped into four classes using cluster analysis The first class had the highest Trans anetole amount that these species were related to temperate regions and the second class had high limonene so that in this class limonene amount showed negative correlation to Trans anetole Results of variance analysis of morphological data totally results of morphological and molecular data indicated that fennel accessions were categorized in different classes according their geographical location Results for self and out pollination traits in fennel indicates self pollination ranged from 2 Paveh to 9 Isfahan cluster analysis of morphological traits classified 15 fennel accessions into four classes so that population located in each class had high similarity Keywords Fennel SRAP GC MS Principle Coordinate Analysis Trans anetole
