Metabolic engineering of mevalonate pathway for production of linalool in the yeast Saccharomyces cerevisiae Nasim Reshadi nejad N reshadinejad@ag iut ac ir Date of submission January Department of Agricultural Biotechonolgy Isfahan University of Technology Isfahan Iran Degree M Sc Language Farsi A Shahpiri Ph D Assist Professor Supervisor a shahpiri@cc iut ac ir M Asadollahi Ph D Assist Professor Supervisor masadollahi@yahoo com Abstract The monoterpenoids are the simplest class of terpenoids containing only carbon atoms two isoprene units which constitute the major components of the plant derived essential oils Most members of this family of natural products are constructed by the monoterpene synthases cyclases that catalyze the conversion of geranyl diphosphate the universal acyclic C intermediate of isoprenoid biosynthesis to the cyclic parents of the various monoterpene skeletal types In the past most studies focused on creating specific heterologous pathways of monoterpenoids in Escherichia coli but recently eukaryotic microorganisms such as yeasts like Saccharomyces cerevisiae has received attentions as a more suitable and preferred host for expression of functional eukaryotic gene products due to many specific characteristics Saccharomyces cerevisiae cannot produce and excrete monoterpenes efficiently mainly because of their lack of monoterpene synthases By metabolic engineering we have modified the isoprenoid biosynthesis pathway in a yeast strain of S cerevisiae expressing the lavendula angostifolia linalool synthase gene an enzyme that uses geranyl pyrophosphate which is produced via mevalonate pathway as a substrate and catalyzes the formation of linalool an acyclic monoterpene found in the floral scents of many plants which has some application in treatment of breast cancer and also leukemia beside its application in industry linalool synthase LIS gene from lavendula angostifolia targeted to different expression vectors of S cerevisiae under the control of four constitutive promoters four constitutive promoters of different strength derived from the genes encoding cytochrome c oxidase CYC alcohol dehydrogenase ADH translation elongation factor TEF and and glyceraldehyde phosphate dehydrogenase GPD These expression vectors then was transformed to yeast S cerevisiae Presence of the LIS gene was confirmed by PCR analysis on genomic DNA The expression of active linalool synthase gene construct in the transgenic yeasts was assessed by measuring linalool emission using solid phase micro extraction SPME GC MS measurements The results obtained with up to g L linalool in the recombinant yeasts The expression system is also comprised of cassettes composed of mentioned promoters flanked by the CYC terminator and has been cleared that the expression rate of linalool synthase in expression vectors with GPD promoter exeed others and then we had more expression in TEF and then ADH promoters We couldn t evaluate any linalool expression in yeasts having expression vectors controlling by CYC promoters PDF created with pdfFactory trial version www pdffactory com
