Micropropagation of Sumac RhuscoriariaL and Detection of Somaclonal Variation in Regenerated Plants Azra Arab azra arab@ag iut ac ir December 14 2013 Department of Biotechnology Isfahan University of Technology Isfahan Iran Degree M Sc Language Farsi M Talebi Assist Prof Supervisor mtalebi@cc iut ac ir B E Sayed Tabatabaei Prof Supervisor sayedt@cc iut ac ir Abstract The medicinal plant sumac Rhus coriaria L Anacardiaceae is a native plant in Iran and has a long history in traditional medicine The antioxidant properties reported in many Rhus species has a role in delaying of lipid peroxidation process in oil and fat foods therefore is recommended for diabetic patients The sumac proliferates by graft and cutting and its proliferation meets to problem through seed because lack of seeds viability of this species and different treatments is not capable to germination Plant tissue culture is a powerful tool for prevent of this species extinction and its proliferation Moreover investigation of some important compounds and antioxidant activity of sumac seems to be essential In this study micropropagation of Iranian sumac performed using the best tissue culture medium and concentration of plant growth regulators The best growth and branching obtained in MS medium containing 1mg l BAP 0 1mg l NAA and 1g l GA3 with 1 5g l activated charcoal in the period of 25 30 days The best rooting obtained in MS medium supplemented with 1mg l BAP 1mg l NAA 1mg l ascorbic acid and 1 5 g l activated charcoal in the period of 14 20 days Application of activated charcoal due to the high amount of phenol in the sumac was necessary in tissue culture at all phases Because of the importance the maintaining genetic uniformity on regenerated plants compared to maternal plants somaclonal variation was examined by 10 SRAP primers The results showed that the somaclonal variation is low and the regenerated plants have not the significant differences to maternal plants The highest band number and polymorphism 38 4 observed in M1E6 primer combination The plants regenerated from node explant therefore low differences appeared are presumably related to structural differences in gene production and biological activities and it have not the significant phenotypic effect Evaluation of chlorophylls and carotenoids showed that there is a significant difference among the three growing periods in amounts of chlorophyll a chlorophyll b total chlorophyll and carotenoids The leaves and fruits are red in the end of sumac growth season therefore observed carotenoid amount in this season was higher in compared to chlorophyll The total amount of phenolic compounds obtained 95 04 mg gDW in the standard curve which according to conducted studies is representing a large amount of phenol in sumac Antioxidant activity increased significantly in all four concentrations of 50 100 300 and 500 ppm as for BHT standard This activity was compared to the high amount of phenol in sumac In amounts of anthocyanins and flavonoids also observed a significant difference among sampling time and different amount of the wavelength p 0 05 In general the changes is different significantly in flavonoid compounds amount compared to variations in anthocyanin compounds amount which anthocyanin amount was more in the end of growth season and it created red color in leaves In conclusion biotechnological methods such as micropropagation and genetic potential of medicinal plants in producing of secondary metabolites can be used as efficient and commercial methods in sumac Keywords Rhuscoriaria tissue culture Somaclonal variation Antioxidant Pigment
