Synthesis and Characterization of Mono and Tri nuclear Pd II Complexes with 4 Chlorobenzoxime and Benzophenone Oxime Ligands Studies of Their Interactions with DNA and BSA Somayeh Askari Barzani s asgari@ch iut ac ir Date of submission 2015 Department of chemistry Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Language FarsiSupervisor Kazem Karami Karami@cc iut ac irAbstractIn this thesis at the first the ligand of 4 chlorobenzoxime was prepared It was synthesized bymixture of 4 chlorobenzaldehyd sodium hydroxide and hydroxylamin hydrochloride in water Benzophenone oxime was synthesized by mixture of benzophenone hydroxylamin hydrochlorideand sodium hydroxide at presence of hydrochloride acid in ethanol Then mono and tri nuclearcomplexes were synthesized respectively 1 Pd3 O N C6H4C Cl NO 4 6 2 Pd3 PPh3 6 3 Pd O N C6H5 2C NO Me3Py The complexes were identified and characterized by FT IR spectroscopy NMR spectroscopy andalso complex 1 was characterized by elemental analysis and X ray crystallography In the second chapter of this thesis the biological activity of 1 and 3 has been evaluated byexamining their ability to bind to Calf thymus DNA CT DNA with UV Vis spectroscopy and CD circular dichroism spectra The results suggested that both complexes interact with CT DNAthrough the groove mode and follow the binding affinity order of Moreover to explore thepotential medicinal value of the new complexes binding interaction of 1 and 3 with bovine serumalbumin BSA was studied at normal physiological conditions using fluorescence and UV Visspectral techniques The reactivity toward protein BSA revealed that the quenching of BSAfluorescence by the complexes The reactivity towards protein BSA revealed that the quenching ofBSA fluorescence by the two complexes are static quenching and complex 3 exhibits a higherBSA binding ability than that of complex 1 Keywords Tri nuclear complexes Groove Static quenching CT DNA binding BSA binding
