111 Taxonomic Study of Ustilaginales Fungi on Poaceae Family in Markazi and Lorestan Provinces Reza Raghebi raghebi r@yahoo com June 15 2015 Department of Plant Protection Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language FarsiSupervisors B Sharifnabi Sharifna@cc iut ac irAbstractThe family Poaceae are annual grasses often with rhizome or stolons and alternate narrow and linear leaveshaving pods in two rows and the covered stems Cereals including wheat barley rice rye oats maize sorghumand millet are the most important plants of 70 world food supply Several plant pathogens attack cereals whichdiffer in geographical distribution damage and attacked developing organs in different environmentalconditions Smut fungi are the most important fungal diseases that often have economic damage on host plants and effect on the growth and reproduction of plant tissues and at the site of infection produces a mass of darkspores called teliospores The spores occur in host tissue as sori and produce as spore mass Smut fungi arebelong to Basidiomycota and class Ustilaginomycetes In this study sampling was done on domestic and wildgramineous plants in Markazi and Lorestan provinces during 2013 2014 The host plants were identified upto species level The morphological characteristics and germination type of teliospore were analyzed using thecurrent identification keys The morphological characters were analyzed using light and fluorescentmicroscopy According to the host teliospore morphology and germination type of teliospores eight species ofUstilago two of Sporisorium and two of Tilletia were identified The results of the morphological and teliosporegermination type analysis could not discriminate the species differentiation The results have shown teliosporevariation in size and shape Teliospore measurements revealed that larger teliospores are belong to Tilletia andsmaller ones to Usitlago and Sporisorium species After DNA extraction amplification of specific regions ofDNA in U hordei Ar7 U nuda LO43 and U tritici LO42 using primers ITS 5 4 rpb1 and tubulinwas done ITS 5 4 primers include regions of ITS1 and ITS2 5 8S sequence and a part of 18S and 28Ssequences Phylogenetic tree for each gene in three isolates was drawn by Neighobor joining order The resultsof the sequencing of these genes ITS rpb1 and tubulin showed that using these genes it s possible todifferentiate these species from each other The U nuda isolate was shown close relationship in compare to U tritici and U hordei In conclusion Differentiation of U nuda and U hordei on barley using morphologicaland host symptoms characters could be done due to closely morphological characters differentiation of U nuda and U tritici on barley and wheat could not be done easily application of each three genes coulddifferentiate the Ustilago species on wheat and barley Keywords Cereals Smut Morphological and molecular analysis Ustilago nuda Ustilago hordei Ustilagotritici Phylogenetic tree
