66 Analysis of Genetic Diversity in Some Pomegranate Genotype Using CAPS and dCAPS Markers Ali Kiani ali kiani8269@gmail com ali kiani1@ag iut ac ir April 3 2016 Department of Agricultural Biotechnology Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language FarsiSupervisors C Ghobadi Cyrus@cc iut ac irB E Sayed Tabatabaei sayedt@cc iut ac irAbstractPomegranate Punica granatum an ancient fruit and known as a heavenly fruit with deep roots inMediterranean and Near Eastern including Iranian cultures which has been cultivated more than twothousand years Although the origin and home of pomegranate is not yet completely known historicalobservations and opinion of most of scientists testify to the point that Iran was the major origin ofpomegranate and it has distributed to other regions of world since it grows in diverse climatic condition Dueto importance of pomegranate and also since Iran is one of biggest producers of this fruit so besides necessityin preserving this natural asset it seems also necessary to pay attention to more research in order to knownative pomegranates and classify them Currently common methods for identification of pomegranate linesare based on appearance of them including color of fruit skin color of seeds taste of seeds and size of fruit Owing to changeability of these traits by climate and also change in name of genotypes with geographicalmovement using these methods to identify the lines is hard and useless Therefore it is necessary to usemarkers which are not influenced by climate and also they contain sufficient and required factors Geneticanalysis can be a strong method for identification of lines measuring detailed genetic relations and drawingphylogenetic tree Among molecular markers CAPS or PCR RFLP and dCAPS with high repeatability stable amplification dominance not usage of complicated facilities and the specificity of their primersaccording to the target gene are of high efficiency The genes being used in this research are MYB and DFR which have been selected due to their high importance in anthocyanin biosynthesis pathway Of the fiveenzymes used in CAPS markers and three pairs of primers along with three enzymes used in dCAPS markers 19 bands were got among which 13 bands created polymorphism and the percentage of polymorphism was68 42 Average Polymorphic Information Content PIC and genetic diversity were 0 41 and 0 35 respectively In order to clustering the PGMA algorithm was used and then Bootstrap test was performed tomeasure the confidence level of drawn tree By clustering about 59 Iranian genotypes were separated andgrouped based on their cultivation type Wild and domesticated genotypes were separated in distinct groups Studied pomegranate lines in this research were classified for better analysis intra and inter groups accordingto their geographical region and cultivation type According to results of AMOVA analysis althoughdiversity was higher intra populations than inter populations difference between populations was completelysignificant p 0 001 Although results of this study showed suitable polymorphism level between 59 linesvia two MYB and DFR genes the resultant genetic diversity with CAPS and dCAPS markers in these twogenes was less than observed morphological diversity Keywords pomegranate CAPS and dCAPS markers genetic diversity AMOVA
