A New Approach in Drug Delivery Synthesis of Nanohydrid Composites with Controlled Drug Release through a Porous Inorganic Coating Mostafa Safari Mostafa safari@ch iut ac ir Department of Chemistry Isfahan University of Technology Isfahan 84156 8311 Iran Supervisor Prof M Ghiaci mghiaci@cc iut ac irDegree PhD Language PersianAbstractIn this thesis three different drug delivery systems were synthesized by following idea ofporous inorganic coating of titanium dioxide TiO2 to reduce speed and control of drugrelease behavior Two of these systems were related to the controlled drug deliverysystems which include Dopamine Chitosan@TiO2 and Insulin Zirconiumphosphate@TiO2 In the former system dopamine first was dispersed in polymeric matrixof chitosan by dissolution evaporation of solvent water and then was coated by TiO2 bysol gel method In the latter system insulin after intercalation between zirconiumphosphate layers like former system was coated by TiO2 Third drug delivery system i e Fe3O4 Metformin Chitosan@TiO2 PEG is categorized under targeted drug deliverysystems In this system first metformin chitosan and Fe3O4 MNPs were mixed bydissolution evaporation of solvent water and then were coated one pot by TiO2 andpolyethylene glycol PEG by formation of covalent bonds between TiO2 and PEG Aftercharacterization of synthesized nanohybrid composites by different techniques such as FT IR XRD DSC TGA BET FE SEM VSM investigation of drug releases of themwere measured by UV visible determination and electrochemical method DPV only firstsystem Drug and coating values were optimized in all three systems Drug release indifferent pHs 1 2 5 4 7 4 were also measured and interesting results were obtained Themain innovation of this work is introduce a porous inorganic coating around the drugdelivery system which improved drug release time from 10 min for first and third systemsand 30 min for second system before coating to over 16 h after coating As well as easyincorporation of drug polymeric matrix and Fe3O4 MNPs and simple procedure to coatingwith PEG third system were other important innovations of these projects Results oftwo first projects could be applied to inject insulin and dopamine contained drug carriersystems with fully controlled release so that intervals of drug injections increasesignificantly and minimizes the need for frequent injections of the drugs Also byimproving the method presented in this study it is possible to achieve an easy way toconvert administration of insulin or other injectable drugs from injection to oral Keywords nanohybrid composite drug release controlled drug delivery targeted drugdelivery dopamine insulin metformin
