Synthesis of Peptide Antibody Complex Nanostructures from Self Assembly of Diphenylalanine PeptidesAbstractNewly self assembly process is considered as a simple and practical way to use some of the material inbiosensors and drug delivery Self assembly process is a process that lead to the emergence of spontaneousand controlled orderly molecular structures in two or three dimensions One of these self assembled materialsis peptide D phenylalanine FF Because of specific reactivity of antibodies to their antigens in the production of bio sensors they have beenused regularly For this purpose the antibodies should be established on the electrode sensing device If theyhad been established physically it is possible that active sites which react with anti female antibodies beunavailable So regular fixation of antibody Anti CD3 FITC on the surfaces of gold silicon and graphitecrystal by using self assembly of peptide D phenylalanine on these surfaces is the subject of this study First we examined the self assembly of peptides FF with diluted concentrations 2 mg per ml andconcentrated 100 mg per ml of solvent HFP on the surfaces of gold silicon and graphite and examined thestability of self assembled peptide structures which formed on the surfaces to solvent by placing them insidethe buffer PBS for 2 hours Also in addition to gold level of concentration we examined the impact of thepresence or absence of 3 and 4 thiol linker toluene on peptide s self assembly and stability To check self assembly and stability of peptides on the surfaces we used SEM analysis and obtained images indicate thatregular formation of self assembled peptide structures with acceptable surface increasing and good stabilityagainst solvents Then by using the FTIR analysis we discussed establishment of the linker peptide on thesurface and junction of thiol group to the gold surface that results indicate the presence of the peptide on thesurface and junction to the gold surface by using thiol linker group In the second stage after activation ofantibody by using EDC and adding it to the peptide by using fluorescence microscopy and FTIR analysis we investigated the activation and the presence of antibodies to the peptide that results indicate establishmentof antibodies on peptide In the final stage of the cycle by using the potentiostat device and cyclic voltammetry analysis CV wediscussed the stabilization of peptide and antibody on the gold surface s electrode and its efficiency inelectrochemical processes for their use in biosensors With the addition of concentrated peptidefunctionalized with thiol linker to the gold surface maximum intensity decreased from 56 to 10 nano ampscurrent through it With the addition of diluted peptide functionalized with thiol linker to the gold surface maximum intensity decreased from 56 to 40 nano amps current through it These changes indicate that thereis a vertical self assembly of peptide concentration functionalized with thiol linker and horizontal self assembly of peptide diluted functionalized with thiol linker on the surface of gold with the addition ofantibodies to the self assembled peptide concentration maximum intensity decreased from 10 to 7 nano amps current through it which is 30 per cent change with the addition of antibodies to the self assembledpeptide diluted maximum intensity decreased from 40 to 15 nano amps current through it which is 62 5 percent change This is an expression of peptide antibodies conjunction that due to greater resistance in thepeptide concentration with the addition of antibody resistance to that we have less decreased currentcompared to peptide diluted Key words peptide Di L phenylalanine self assembly antibody biosensor
