75 Assessment of Genetic Diversity within Prangos ferulacea L Lindl Populations Using SCoT and SRAP Molecular Markers and Total Phenolic Content Farideh Rahimi Chermahini faride rahimi1@ag iut ac ir January 10 2018 Department of Agricultural Biotechnology Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Language Farsi Supervisors M Talebi mtalebi@cc iut ac ir M Rahimmalek mrahimmalek@cc iut ac irAbstractPrangos ferulacea L Lindl from Apiaceae family and considered as an important medicinal herb isprescribed as a stomatch laxative stomach acid anti flatulence soothing nerves anti inflammatory andantiviral medication in traditional medicine The origin of P ferulacea is in the Iran and parts of Turkey Italy Kazakhstan and Syria Despite the medicinal importance of this plant molecular studies have notbeen done to assess the genetic diversity of P ferulacea In this study assessment of genetic diversitywithin P ferulacea species was done using SCoT and SRAP molecular markers which target expressibleregions of the genome Nine SCoT primers and nine SRAP primer combinations were applied to assess thegenetic diversity of 58 genotypes and amplified 217 and 200 bands which showed 93 59 and 92 46 ofpolymorphism respectively In order to investigate the primers efficiency the discerning of genotypes polymorphic information content PIC was calculated and it was 0 42 for SCoT primers and was 0 417for SRAP primer combinations The dendograms generated from Jaccard s similarity matrix and theUPGMA method revealed genetic relationships among the P ferulacea genotypes The highest copheneticcorrelation coefficient of SCoT and SRAP analyses were 0 51 and 0 53 respectively which relativelyindicated the separation of populations The analysis also revealed a relatively acceptable differentiation inallele frequencies that was equal to Fst 0 015 in SCoT analysis and it was equal to Fst 0 022 in SRAPresults which illustrated high genetic diversity and gene flows among populations High percent ofdiversity among populations indicates a rich resource of germplasm for breeding plants Besides theassessment of population structure demonstrated high rate of gene flows and mixture of populations and itseems that classification of P ferulacea genotypes does not related to their geographical origin Moreover the total phenol composition was assessed using spectrophotometry and folin ciocalteu method and arelatively extent variety 0 715 0 723 milligram of tannic acid per gram of dry weight of plant was shownamong populations In addition HPLC test using standards of phenolic compositions defined nine indexcompositions related to seven geographical regions consisting of galic acid cholorogenic acid ferolicacid coffeic acid pcoumaric acid luteolin rutin quercetin and apigenin The result of HPLC illustrated agreat variety of phenolic content 0 414 38 957 mg dry weight of plant among P ferulacea samples Inconclusion SCoT and SRAP molecular markers have good potential for assessment of genetic diversity ofP ferulacea genotypes but they could not separate populations based on their geographical regions Besides assessment of total fenol content of ferulacea s genotypes can provide more comprehensiveinsights regards to this valuable plant Keywords Prangos ferulacea Genetic diversity SCoT SRAP Total phenolic content
