Production and evaluation of core shell PLGA Gelatin fibers loaded withciprofloxacin hydrochloride and gabapentin for neuropathic diabetic wound healing Mahsa Mostofizadeh m mostofi@tx iut ac ir Date of Submission 14 01 2018 Department of Textile Engineering Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language FarsiSupervisor Dr Lale Ghasemi Mobarake laleh ghasemi@cc iut ac irAbstract Diabetes mellitus is one of the most common chronic medical conditions affecting over 100 millionpeople world wide Diabetic neuropathic pain DNP has always been a challenging complication of diabetesmellitus DNP is severe and intractable and affects about 10 26 of millions of patients with diabetes DNPmanifests as burning excruciating stabbing infections or intractable type of pain or presents with tinglingor numbness In this research the non woven wound dressing with core shell structured fibers was preparedby coaxial electrospinning Gelatin was electrospun as the fiber s core whereas poly lactic co glycolic acid was fabricated into the shell in order to healing the neuropathic diabetic wound Simultaneously gabapentinas analgesic drug was loaded in the shell whereas ciprofloxcacin hydrocholoride as antibiotic drug wasencapsulated in the core In order to produce optimum core shell fibers the concentration of each component wasdetermined The core shell structure of fibers was investigated using TEM and Fluorescence microscope Morphology of core shell fibers were studied by SEM Fourier transform inferared FTIR Differential scanningcalorimetry DSC Thermal gravimetric analysis TGA X ray diffraction XRD were used for characterizationof core shell fibers Properties of core shell fibers after and before adding drugs were evaluated by contact anglemeasurement biodegradation test and tensile measurement analysis Resulting from the fiber s core shellstructure cirofloxcacin hydrocholoride released from the core and gabapentin present in the shell can endow thedressing both painkiller and anti bacteria ability simultaneously which can greatly enhance the dressing s clinicaltherapeutic effect The dressing s anti bacteria ability against Staphylococcus aureus and Escherichia coli wasproved by in vitro anti bacteria test The in vitro drug release test showed the sustainable release of ciprofloxcacinhydrocholoride and burst release of gabapentin within 64 days In addition to it was shown that the concentrationof polymer and drugs influence significantly on profile release In conclusion the fabricated core shell fibersdressing possesses multiple functions to benefit wound healing and shows promising potential for clinicalapplication Keywords diabetic neuropathy coaxial electrospinning core shell fibers gelatin poly lactic co glycolic acid
