Identification of Rhychosporium species and determination of genetic structure of populations of barley scald pathogen in Iran Elaheh Seifollahi e seifollahi@ag iut ac ir December 5 2018 Department of Plant Protection Isfahan University of Technology Isfahan 84156 8311 Iran1st Supervisor Bahram Sharifnabi sharifna@cc iut ac ir2nd Supervisor Mohammad Javan Nikkhah jnikkhah@ut ac ir Professor in Plant Pathology Department of Plant Protection College of Agriculture Isfahan Universityof Technology Professor in Mycology and Fungal Plant Pathology Department of Plant Protection College ofAgriculture and Natural Resources University of TehranAbstractDifferent species of Rhynchosporium including R commune R secalis R agropyri R orthosporum and R lolii cause scald disease or leaf blotch on barley and some grasseswhich reduces the quantity and quality of the yeild In this study 204 isolates frombarley and uncultivated grasses were collected from Gorgan Miandoab Eyvan andBaghmalek Morphological and phylogenetic analyses of two ITS and beta tubulin lociwere used to identify Rhynchosporium species and their host ranges in Iran Conidia of68 isolates from barley and wild grasses were measured for morphologicalidentification The mean of total conidial lengths was 16 79 with standard error of themean 0 04 and varied between 15 00 22 53 m among different isolates The mean oftheir conidial widths were 3 06 with standard error of the mean 0 006 and variedbetween 2 73 3 50 m among different isolates Despite the large variation observedfor conidial dimensions the phylogeny constructed from the ITS rDNA region and tubulin gene revealed that all isolates from barley and wild grasses in Iran belong to R commune R commune is reported for the first time from Hordeum murinum ssp glaucum H vulgare ssp spontaneum and Lolium multiflorum from Iran This study isalso a first report of Avena sativa matrix nova as a new host of R commune Crossinoculation of R commune isolates from all hosts uncultivated grasses and cultivatedbarley showed that uncultivated grasses could be as an inoculum sources for cultivated 1
