Isolation and characterization of metal solubilizing bacteria from contaminated soils and their bioaugmaentation effects on phytoavailability of zinc and lead Zahra Yahaghi z yahaghi@ag iut ac ir January 20 2019 Department of Soil Science Isfahan University of Technology Isfahan 84156 83111 Iran1st Supervisor Mehran Shirvani shirvani@cc iut ac ir2nd Supervisor Farshid Nourbakhsh farshid@cc iut ac ir Professor associate of Soil Science Department of Soil Science College of Agriculture IsfahanUniversity of Technology Professor of Soil Microbial Ecology Department of Soil Science College of Agriculture IsfahanUniversity of TechnologyAbstractThe aim of this study was to isolate and characterize lead Pb and Zinc Zn solubilizingbacteria by plant growth promoting characteristics as well as heavy metal tolerance fromheavy metal contaminated mine soils and to evaluate their inoculation effects on growthand Pb and Zn absorption of plant Only Small fraction of isolated bacterial strains wereable to dissolve the sparingly soluble Pb and Zn minerals Based on 16S rDNA genesequence analysis most of the isolated strains belonged to Bacillus genera A majoritysolubilizing strains were able to produce indole acetic acid and siderophores anddisplayed tolerance to different heavy metals In a study conducted in agar medium Bacillus cereus YSP4 and Bacillus filamentosus YSP110 were the most effectivebacterial strains in stimulating alfalfa seedlings growth and heavy metal accumulation ina Zn and Pb contaminated medium respectively In the experiment conducted oncontaminated soil by inoculation of corn and Indian mustard with selected Pb and Zn solubilizing strains demonstrated that the effect of bacteria on plant growth is depend onplant type In addition selected Pb solubilizing bacteria and the consortium of two Zn solubilizing bacteria were more efficient in enhancing Pb and Zn concentration in plant respectively Moreover the result of this study showed that bacterial inoculation byBrevibacterium frigoritolerans YSP40 is more capable to enhance growth as well as Pbconcentration of corn shoot in the presence of PbCO3 and PbO supplemented sandmedium In the sand culture supplemented by sparingly solubilize Zn compound bacterial inoculation was also efficient in enhancing plant growth and Zn concentrationin shoot of corn Therefore the bacterial strains isolated in this study are promising
