چكيده
بيماري ساق سياه و پوسيدگي نرم سيب زميني از مهم ترين بيماري هاي سيب زميني محسوب مي شوند كه در مزرعه و مراحل پس از برداشت، خسارت قابل توجهي به اين محصول وارد مي كنند. اين بيماري ها توسط گونه هايي از دو جنس باكتريايي Pectobacterium و Dickeya ايجاد مي شوند كه در سال هاي اخير و به دليل افزايش دماي اقليمي، گونه هاي گرمادوست P. parmentieri (Pp)، P. brasiliense (Pbr) و (Dsol) D.solani انتشار بيشتري در مزارع جهان و نيز ايران پيدا كرده اند. به دليل يكسان بودن فرايند توليد غده هاي خوراكي و بذري سيب زميني درايران، احتمال آن وجود دارد كه اين عوامل بيمارگر باكتريايي از مناطق آلوده به ساير نواحي كشت سيب زميني نيز منتقل شوند. بنابراين، كسب اطمينان از عدم آلودگي غده هاي بذري سيب زميني به اين باكتري هاي بيمارگر، براي براي كاهش شيوع بيماري هاي ساق سياه و پوسيدگي نرم در كشور لازم است. به اين لحاظ، دستيابي به يك روش كار آمد و ساده براي رديابي سريع Pp، Pbr و Dsol در غده هاي بذري سيب زميني، به عنوان هدف اصلي اين پژوهش در نظر گرفته شد . به اين منظور، ابتدا دو جدايه P7 و P60 از Pp، L4 و P63 از Pbr و نيز Dh12 و D2222 از Dsol براي انجام آزمايشات اوليه انتخاب شدند. هيچكدام از اين جدايه ها در ℃5 توانايي ايجاد پوسيدگي نرم در برشهاي غده سيب زميني را نداشتند، ولي علايم بيماري در ℃28 براي تمام جدايه ها شديد بود. پيشرفت لهيدگي جدايه هاي Dsol در دماي ℃35 نسبت به جدايه هاي Pp وPbr بيشتر ارزيابي شد. پس از به كارگيري روش هاي استاندارد باكتري شناسي و اطمينان از همساني ويژگي هاي مورفوژيكي، بيماري زايي، فيزيولوژيكي و بيوشيميايي جدايه هاي هرگونه، سه جدايه P60 از Pp، L4 از Pbr و نيز Dh12 از Dsol براي انجام آزمون هاي رديابي مولكولي باكتري به روش Multiplex-PCR انتخاب شدند. پس از استخراج DNA از جدايه ها به دو روش CTAB و جوشانيدن ، ابتدا تكثير باند مورد نظر DNA اين باكتري ها به ترتيب با جفت آغازگرهاي اختصاصي Pw7011F/R براي Pp (bp 140)، BR1F/R براي Pbr (bp 322) و DSF1/DSR2 (bp 895) تاييد شد. در مراحل بعد و طي آزمون هاي مكرر، با استفاده از روش رقيق كردن متوالي DNA جدايه ها و جفت آغازگرهاي اختصاصي و نيز تغيير دماي اتصال آغازگرها در آزمون هاي Gradient-PCR ، غلظت بهينه DNA، غلظت جفت آغازگرها و دماي اتصال واكنش براي انجام آزمون هاي Duplex-PCR و Multiplex-PCR بررسي گرديد. بر اساس نتايج بدست آمده، غلظت 40ng/µl از DNA هر باكتري، به كارگيري غلظت pmol 10 از هر آغازگر، افزودن 25mM از Mg Cl2 به عنوان مقادير بهينه هر مخلوط واكنش 20 ميكروليتري و استفاده از دماي اتصال 62℃ و دماي گسترش 65℃ در انجام هر واكنش ، براي تكثير قطعات DNA مورد انتظار آزمون هاي PCR، Duplex PCR و Triplex PCR تعيين شد. در تعيين كارآيي آزمون هاي سه گانه PCR براي رديابي باكتري هاي Pp، Pbr و Dsol از غده هاي آلوده، ابتدا، سوسپانسيوني از هر جدايه به صورت انفرادي، مخلوط دوتايي و مخلوط سه تايي در غده هاي جداگانه مايه زني شدند. پس از بروز لهيدگي در بافت غده ها، يك گرم از بافت لهيده هر تيمار براي استخراج DNA بكار رفت و سپس واكنش هاي PCR، Duplex PCR و Triplex PCR براي هر تيمار انجام گرفت. نتايج نشان داد كه روش هاي بهينه سازي شده Multiplex PCR مي تواند بصورت دقيق و با با كيفيتي مناسب باكتري هاي هدف را در غده هاي آلوده شناسايي كند. در مجموع، مي توان گفت امكان رديابي منفرد و يا همزمان سه جدايه Pp، Pbr و Dsol از غده هاي سيب زميني با روش بهينه سازي شده Multiplex-PCR در اين پژوهش موفقيت آميز بود. بنظر مي رسد روش هاي Multiplex-PCR توسعه يافته در اين پژوهش براي غربالگري غده ها ي سيب زميني از نظر آلودگي به باكتريهاي Pp، Pbr و Dsol به لحاظ اهداف قرنطينه اي و نيز صدور گواهي سلامت بذر مفيد واقع شود.

Abstract
Potato blackleg and soft rot are among the most important diseases of potato, causing significant losses to this crop both in the field and during the post-harvest period. These diseases are caused by species belonging to two bacterial genera, Pectobacterium and Dickeya, and in recent years — largely due to rising climatic temperatures — the thermophilic (heat-tolerant) species P. parmentieri (Pp), P. brasiliense (Pbr), and D. solani (Dsol) have become more widespread in potato fields worldwide, including in Iran. Because the production processes for table (ware) and seed potato tubers in Iran are not separated, there is a risk that these bacterial pathogens could be transmitted from infected regions to other potato-growing areas of the country. Ensuring that seed potato tubers are free of these pathogenic bacteria is therefore essential for reducing the incidence of blackleg and soft rot diseases nationwide. Accordingly, the main objective of this study was to develop a rapid and simple method for detecting Pp, Pbr, and Dsol in seed potato tubers.
For this purpose, two isolates each of Pp (P7 and P60), Pbr (L4 and P63), and Dsol (Dh12 and D2222) were initially selected for preliminary experiments. None of these isolates was able to induce soft rot in potato tuber slices at 5°C, whereas disease symptoms were severe for all isolates at 28°C. Rot progression of the Dsol isolates at 35°C was found to be greater than that of the Pp and Pbr isolates. After confirming, through standard bacteriological methods, that the morphological, pathogenicity, physiological, and biochemical characteristics were consistent among isolates of each species, three representative isolates — P7 (Pp), L4 (Pbr), and Dh12 (Dsol) — were selected for molecular detection by multiplex PCR.
Following DNA extraction from the isolates by both the CTAB method and the boiling method, amplification of the target DNA fragments was first confirmed using the species-specific primer pairs Pw7011F/R for Pp (140 bp), BR1F/R for Pbr (322 bp), and DSF1/DSR2 for Dsol (895 bp). In subsequent steps, through repeated trials involving serial dilution of the isolatesʹ DNA with the specific primer pairs, as well as Gradient-PCR assays to optimize primer annealing temperature, the optimal DNA concentration, primer concentration, and annealing temperature for Duplex-PCR and Multiplex-PCR reactions were determined. Based on the results obtained, the optimal conditions for each 20 µl reaction mixture were established as: 40 ng/µl of DNA from each bacterium, 10 pmol of each primer, and 25 mM MgCl₂, with an annealing temperature of 62°C and an extension temperature of 65°C, for amplification of the expected DNA fragments in the PCR, duplex-PCR, and triplex-PCR assays.
To evaluate the efficiency of the three PCR-based assays for detecting Pp, Pbr, and Dsol in infected tubers, bacterial suspensions of each isolate — individually, in binary mixtures, and in a ternary mixture — were inoculated into separate tubers. After the onset of soft rot symptoms in the tuber tissue, one gram of macerated tissue from each treatment was used for DNA extraction, followed by PCR, Duplex-PCR, and Triplex-PCR reactions for each treatment. The results showed that the optimized Multiplex-PCR protocols could accurately and reliably identify the target bacteria in infected tubers. Overall, it can be concluded that the individual or simultaneous detection of the Pp, Pbr, and Dsol isolates from potato tubers was successfully achieved using the multiplex-PCR method optimized in this study. It appears that the multiplex-PCR protocols developed in this research could be useful for screening potato tubers for contamination with Pp, Pbr, and Dsol for quarantine purposes as well as for seed health certification.
