Abstract This study was conducted to saturate the linkage map of wheat using theAFLP markers and also mapping the QTLs for some important quantitative traits suchas plant height heading date date to maturity grain yield and yield components Adoubled haploid population derived from crossing of two cultivars Fukuho kumogiand Oligo culm was used The parental lines and 157 lines were evaluated in arandomized complete block design with three replications at the research farm ofIsfahan University of Technology in two years of 1382 and 1383 The analysis ofvariance in each year and combining the data over years for the agronomic andmorphological traits showed significant differences among the genotypes for all of thetraits The estimation of genetic and phenotypic coefficients of variations indicatedthat there was more genetic variation for plant height peduncle length number ofgrains and grain weight per spike spikes per m2 spike length and grain yield Theestimation of heritability for plant height heading date number of grains per spike grain weight per spike and date to maturity were 98 0 88 7 87 3 85 9 and80 3 respectively The AFLP analysis using 11 primer combinations of PstI ANN MseI CNN amplified 66 polymorphic bands between the parents but 48 markerswere assigned to the wheat linkage groups The mean of amplified polymorphic bandswas 6 per each primer combination The primer combinations of PstI AAT MseI CCC PstI AAT MseI CAC and PstI AGA MseI CCC each with 9 bands hadhigher polymorphic bands In this study 41 of AFLP markers assigned to thelinkage groups of genome A 43 to genome B and 12 to genome D The results ofmapping QTLs for the studied traits indicated the expression of the permanent QTLswith main effects for plant height peduncle length spike length spikes per m2 number of grains per spike grain weight per spike and heading date The resultsshowed two QTLs with main effects were located in the expected location of Rht1 andRht2 genes on the chromosomes 4B and 4D for plant height that explained 60 3 and61 1 of phenotypic variation of this trait in 1382 and 1383 respectively In theinterval of 29 0 to 34 0 centimorgan from the beginning of chromosome 2D and closeto the Xcfd53 and Xgwm261 markers the QTLs for awn length R2 11 0 spikelength R2 31 0 plant height R2 10 8 peduncle length three QTLs with R2 7 5 date to maturity R2 11 0 pollination date R2 10 7 harvest index R2 12 4 grain yield two QTLs with R2 6 5 spikelete compactness R2 18 number of grains per spike R2 3 5 and grain weight per spike R2 6 4 wereidentified There was a main QTL for grain yield between the markers of Xpsp2999and Xwmc333 on chromosome 6B 3 centimorgan far from Xwmc333 which was alsoin the vicinity of wheat strip rust QTL One of the other main QTLs for grain yieldwas mapped on chromosome 6A2 and in the distance of less than 50 centimorganfrom powdery mildew QTL which is close to the Xgwm427 marker The QTLslocated in vicinity of Xpsp2999 and Xgwm261 markers had much contribution to thephenotypic variation for the number of grains per spike grain weight per spike grainyield spike number per m2 plant height and spike length
