AbstractIran is one of the centers of genetic diversity for cultivated and wild safflower species Someof the wild species such as Carthamus oxyacantha having distribution and adaptability withdifferent climate conditions could be used in safflower breeding programs In this study populations of wild safflower C oxyacantha were collected from western central andsouthern regions of Iran to investigate their genetic variation for quantity and quality of seedoil agro morphologic traits and ISSR markers The effect of seed coat color on resistance tosafflower fly Acanthiophilus helianthi was also evaluated Furthermore the inheritance andpossibility of transferring seed coat color and other useful genes from wild species tocultivated one via interspecific hybridization was studied The mechanism of plant resistanceto safflower fly and effective bio chemical compounds involved in the resistance were alsoinvestigated Light and electron microscope were used to observe the color ingredientsaccumulation part in the seed coat Finally the effect of seed color on seed yield and otheragronomic traits of safflower and fly resistance were studied in F3 generation derived from theinterspecific hybridization Evaluation of genetic diversity among wild safflower accessionsusing morphologic traits and ISSR markers showed that there were considerable variationamong the wild accession and based on clustering they were separated according to theirdifferent geographical regions The results showed that some wild accessions had appropriateseed oil content and oil quality comparable with the oil extracted from cultivated species Inthe wild safflower germplasm colored seeds brown and black had higher frequency higherseed weight and lower percentage of safflower fly damage compared to the white seeds Higher dead fly larvae in the heads and lower infected heads resulted in lower fly density andseed yield damage in the wild safflower accessions than the cultivated species This mayindicate that wild species are possibly resistant to safflower fly Detection of differentpolyphenolic flavonoid compounds in colored seed coats showed that these compounds mayinvolve in antibiosis of fly larvae Study of F2 and F3 progenies of interspecific cross showedthat two loci with epistatic action may control seed coat color Also evaluation andcomparison of F3 population having white or colored seed coats in the field showed thatplants derived from colored seed had better seedling characteristics and earliness higher yieldand yield components specially the number of heads per plant but lower seed hull percentage Seed coat color had also significant effect on safflower fly damage The number of eggs andpupae in infected heads and fly density on population derived from colored seeds were lowerthan the population derived from white seeds There were considerably higher dead larvae incolored seed populations than the white seeded ones Yield per infected plant was higher incolored seed population than the white seed counterparts thus yield loss percentage due tosafflower fly was lower in colored seed populations These results may show that transferringcolored seed coat to cultivated safflower could render plant resistant to safflower fly damage Key words Antibiosis Polyphenol Safflower fly Seed color Wild safflowerSafflower Carthamus tinctorius L is an important oilseed crop and an essential component
