Sequencing of Some Genes Involved in the Synthesis of Antioxidants and Its Application in Phylogeny Important Genera of Anthemideae Subfamily Monireh Sadeghi m sadeghidastjerdi@ag iut ac ir Date of submission February 25 2012 Department of Agricultural Biotechnology Isfahan University of Technology Isfahan Iran Degree M Sc Language Farsi 1 M Rahimmalek Ph D Assist Prof Supervisor mrahimmalek@cc iut ac ir 2 A Yamchi Ph D Assist Prof Supervisor yam12001@yahoo com 3 B E Sayed Tabatabaei Prof Advisor sayedt@cc iut ac irAbstractAnthemideae is considered as an important subfamily of Asteraceae In this subfamily five genera havehigh economical and medicinal value including Achillea sp Matricaria sp Tanacetum sp Artemisiasp and Santolina sp These plants possess bioactive compounds such as antioxidants Antioxidantsincluding two groups named as enzymatic and non enzymatic Plant antioxidants have fewer side effectscompared to synthetic ones As plants belonging to Anthemideae are rich for antioxidants detection ofgenes involve in synthesis of these compounds is of great importance In this study four crucialantioxidant genes aox2 related to AOX or Alternative oxidase and 3 other genes pal1 pal2 and pal3 which were related to PAL phenylallanin ammonia lyase were detected in five genera of Anthemideae According to Gene bank information and using OLIGO ver 5 0 software specific primers were designedfor these genera aox2 and pal2 had clear amplification while for pal1 and pal3 no bands were observed aox2 primers amplified a 300 bp fragment in all studied genera while a 700 bp fragment could beobserved from amplification of pal2 gene in Artemisia sp Chrysanthemum sp Matricaria sp andAchillea sp genera The amplified fragments were sequenced and aligned with other plants in NCBI database The results obtained by BLAST search in NCBI database confirmed the correction of sequences The results were also confirmed by multiple sequence alignment of these genera with other plants assessment of amino acid sequence in regards to their nucleotide sequence and the presence of the activeregion of PAL2 and AOX2 protein sequences Nucleotide sequence amino acid sequence and activedomains assessment showed that these fragments were corresponding to pal2 and aox2 genes Wecalculated parwise distance based on K2P p distance model and genetic diversity based on transition andtransversion to investigate the intergenus variation among some important genus of Anthemideae Average pairwise distance was also calculated between studied genera and Artemisia annua and othergenera of Asteraceae family Calculating the Pairwise distance based on K2P aox2 0 069 pal2 0 177 and P distance aox2 0 059 pal2 0 156 and genetic diversity based on transition and transversion aox2 14 5 pal2 30 2 indicated that these genera were more conserved based on aox2 gene whilepal2 showed more diversity According to pairwise distance results Matricaria sp and Achillea sp werethe closest genera while Artemisia sp and chrysanthemum sp had the most distance which was inagreement with results of ndhf choloroplastic gene Pal2 region had low diversity in compare to aox2region Dendrogram was plotted using the maximum parsimony for aox2 gene was similar to result whichcreated in cluster analysis with ndhf gene and ITS sequence that can be due to high similarity ofmentioned genus in aox2 gene Keywords Antioxidants Anthemideae aox2 pal2
