81 Effects of Indol 3 Butyric Acid and Putrescine on the Rooting of the Olive Cuttings Somayeh kadkhodaei kadkhodaei somaye@yahoo com 15 april 2012 Department of Horticultural Science Isfahan University of Technology Isfahan 84156 83111 IranDegree M Sc Language FarsiSupervisor Prof M Mobli mobli@cc iut ac irAbstractOlive is a holly evergreen subtropical and long life tree that is native to Mediterranean area The usualmethod of propagation of this tree is semi hardwood cutting The cuttings of olive are more difficult toroot Rooting disability can be efficient in developing of new orchards with appropriate adaptable andacclimated cultivars In this research the effect of two growth regulators indol 3 butyric acid IBA putrescine and their interaction on the root regeneration and subsequent growth of semi hardwoodcuttings of olive were studied In the first experiment which has performed in 1389 three concentrationsof IBA 0 2000 and 4000 mgl 1 and three concentrations of putrescine 0 0 1 and 1 Mm were used on Zard and Roghani cultivars The experiment was carried out as 3 3 2 factorial experiment in acompletely randomized design whit four replications Results showed that the percentage of healthy androoted cuttings were higher in Zard cultivar compared with Roghani Treatment of cuttings with 2000mgl 1 IBA increased total percentage of cutting with callus production and root regeneration rootnumbers and root length per cutting branching and number of branches per cutting compared withcontrol and 4000 mgl 1 IBA Putrescine at the concentrations of 0 1 and 1 mM showed the best resultsregarding percentage of cuttings with callus and root production numbers and length of root per cuttingand new shoot produced on cuttings A significant interaction between IBA and putrescine was observedso that the largest percentage of healthy and rooted cuttings were obtained in combination of 2000 mgl 1IBA with 0 1 and 1 mM putrescine In the second experiment which was done in 1390 only Zard cultivar was used Treatments were combination of 2000 mgl 1 IBA with one of the three putrescineconcentrations of 0 0 1 and 1 mM The experiment was done as a completely randomized design withfour replications Sample of cuttings were taken for measurments in 3 times two three and four monthsafter cutting In this study two levels of 0 1 and 1 mM putrescine increased percentage of cuttings withcallus and root production numbers and length of root per cutting branching and number of shoots percutting compared to control treatment whereas there was no significant differences between these twoconcentrations Also by reviewing the effect of IBA and putrescine along time results revealed that thepercentage of cuttings with callus were higher after two months from cutting and treating with IBA andputrescine compared with three and four months whereas rooted cuttings were higher at three and fourmonths sampeling By the time the number and length of the roots and also number of shoots per cuttingwere increased Overall use of 0 1 and 1 mM putrescine following treatment of cutting with 2000 mgl 1IBA were better treatment so that it can help rooting and following growth of the cuttings for both olivecultivars so it can be used for improving propagation method of this plant Key words Olive indol 3 butyric acid putrescine cutting callus and rooting
