111Application of SSR Markers for Studying Intra and Inter Specific Variations in the Genus Brassica Yousef sharafi sharafiyousef@yahoo com Submitted on September 15 2012 Department of Agronomy and plant Breeding Isfahan University of Technology Isfahan 84156 83111 IranDegree M S c Language FarsiM M Majidi Assist Prof supervisor Majidi@cc iut ac irM H Ehtemam Assist Prof supervisor hehtemam@cc iut ac irAbstractCanola rapeseed is one of the oilseed crops widely cultivated in Iran To ensure efficient rapeseed productionbreeders have aimed to produce high yield and high quality cultivars Wild relatives of Brassica possess a number ofuseful traits such as male sterility resistance to disease and pests tolerance to cold salt and drought conditionswhich could be incorporated into breeding programs Evaluation of genetic diversity and relationship of genomeamong wild and crop plants population is necessary for conservation and useful application of germplasm Toinvestigate genetic diversity and relationship of some Brassica species based on morphological traits SSR markerand botanical characteristics three experiments were conducted during 2010 2012 In the first study geneticdiversity between 36 accessions of six Brassica species was evaluated for morphological traits using multivariatestatistical analysis Results indicated that considerable variation was found between accessions for all traits andbetter performance for B carinata species in respect of seed yield The estimation of broad sense heritability for podlength 1000 seed weight and day to repining was high suggesting contribution of the major genes for controllingthese traits Cluster analysis and principle component analysis PCA could separate species based on morphologicaltraits Stepwise regression for seed yield as independent variable showed that number of branches per plant numberof pods in main stem number of seeds per pod 1000 seed weight and day to ripening explained the majority of thevariation Also results of path analysis coefficient indicated that number of branches per plant number of seeds perpod 1000 seed weight number of pods in main stem and day to ripening had the highest direct effect on seed yieldindicated that these traits can be used for constructing a selection criterion to improve seed yield in rapeseed In thesecond study genetic diversity between 56 accessions of ten species was evaluated using SSR technique From 27primers 127 polymorphic markers were scored to differentiate between accessions Result showed that geneticvariation within accessions of B napus was less than other species Applying UPGMA method of cluster analysisand PCA species were appropriately separated from each other and classified based on their genome similarity However cluster analysis did not coincidence geographical regions The analysis of molecular variance AMOVA based on species showed that majority of the genetic variance 56 was found within the species and less amongthe species In the third study genetic variation of 57 accessions of Brassica genus were evaluated using anatomicaltraits Although analysis of variance for these traits showed high variation among and within species no clearrelationship was observed among the genomes of species Based on the cluster analysis no coincidence was foundbetween anatomical variation with species and geographical regions Results of principle components analysisconfirmed the result of cluster analysis There was not any significant correlation between clustering created fromanatomical data with molecular and morphological ones Also moderate correlation r 0 5 was found betweencluster of morphological and SSR data However SSR marker could much better determine the genetic similarityand diversity of accessions and species indicating the efficiency of this marker for genetic studies Keywords Brassica morphology anatomy molecular cluster analysis phylogeny genetic similarity
