AbstractThere is limited knowledge about cellular and histological responses to endometritis in Iraniandairy cattle farms Neutrophils PMNs migration to the lumen is one of the first uterineresponses to infection so evaluating PMNs proportion in vaginal mucus of postpartum dairycows is useful for determination of uterus inflammation severity Histological changes in uterussuch as denudation of lining epithelium and infiltration of neutrophils in endometrium are causedby endometritis in dairy cattle too Furthermore high progesterone concenration is a risk factorfor endometritis So present study was designed to show comparative cellular and histologicalcharacteristics of healthy and endometritic cow s uterus specially PMNs index in couple ofexperiments In the first experiment genital tracts of 9 healthy and 9 endometritic cows werecollected from slaughterhouse to examine apparent characteristics and tonicity of uterus Middlepart of the body and larger horn of uterus were sampled and stained with H E procedure In thesecond experiment 15 non pregnant holstein cows in second parity were used that showedhealthy ovaries had female calf and were inseminated at least twice after parturition Bloodsamples were collected at days 0 3 and 9 of oestrus cycle and also mucus were sampled at day 0 Mucus samples were air dried on glass slides stained with Pap procedure and then were studiedat magnification 400 by light microscope The apparent uterus characteristics showed non significant differences between control and endometritic groups but tonicity score of uterus inendometritic cows was significantly more than healthy cows p 0 05 Histologically significantincrease occured in endometrium diameter P 0 01 neutrophils infiltration P 0 05 glandsdispersal P 0 05 and percent of fibrotic regions P 0 05 in endometritics cows In addition staining density and cellular solidarity in endometrial glands significantly decreased inendometritic cows in comparison with healthy cows Using cytology for vaginal mucus in dairycows neutrophils population amount of cell changes and bacterial population showedsignificant differences between control and endometritic groups but there were not significantdifferences between Trichomonase population and cell maturatin index The data of progesteroneconcentration did not show significant differences between healthy and endometritic cows atdays 0 3 and 9 of oestrus cycle The results of present study indicated that using cytology inpostpartum dairy cows might be a beneficial procedure for diagnosing of endometritis based onneutrophil and bacterial population Key words endometritis cytology histology dairy cattle
