Genetic Diversity of the Persian Fallow Deer Dama mesopotamica by mtDNA D loop sequence variation Habiballah Haghi hhaghghi@yahoo com Date of Submission 2012 9 15 Department of Natural Resources Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Language FarsiSupervisor Mansoureh Malekian mmalekian@cc iut ac irAbstract The Persian fallow deer Dama mesopotamica is a rare species in the world After presumed extinctionof this species in the wild captive breeding has been used in human controlled environment and introducedto the wild This species has classified as Endangered EN on the IUCN Red List In the current study wehave investigated the genetic diversity and population structure of the Persian fallow deer by analyzingmitochondrial DNA D loop sequence DNA was extracted from bloods and muscle samples of 39 individualscaptured from Iranian populations A pair of primers was designed using the mtDNA D loop sequenceretrieved from National Center for Biological information NCBI The target region was amplified and theresulting products were sequenced The nucleotide composition of the all sequences was investigated whichincluded 25 6 Adenine 28 9 Thymine 26 1 Cytosine and 19 4 Guanine We find no genetic variationat 402bp of mtDNA control region among Iranian populations of the Persian fallow deer The obtainedsequences were used for comparison of Persian fallow deer with sequences of other Cervidae speciesavailable from GenBank A phylogenetic tree was constructed using neighbor joining method with thenumber of replications for bootstrap test was set to 1000 The results showed that the Persian fallow deer andEuropean fallow deer are genetically divergent and distinct species Also the European fallow deer haplotype were not found among Iranian Persian fallow deer population based on the maternal region of D loop usedin this study However more studies using bio parental genes are needed to support this hypothesis AsPersian fallow deer has been illegally hunted in the wild a pair of primers was designed in conserved regionof D loop which amplified a fragment of 588 bp Samples from other suspected species such as jabber Gazella bennettii wild goat Capra aegagrus urial Ovis orientalis cattle and sheep were obtained DNAwas extracted from these samples and used in PCR amplification The result showed that the designedprimers only amplify the Persian fallow deer samples and no other species was amplified using these primers suggesting that these primers can be used for defining these species where identification is required Keywords Persian Fallow Deer Dama mesopotamica Genetic diversity D loop region specific primers
