Assessment of genetic diversity within and among sage Salvia sp species using SRAP marker and total phenolic compositions Zahra Aghaei Jeshvaghani z aghaeijeshvaghani@ag iut ac ir Date of submission Januray 08 2013 Department of Agricultural Biotechnology Isfahan University of Technology Isfahan 84156 83111 Iran Degree M Sc Language Farsi 1 M Talebi Assist Prof Supervisor mtalebi@cc iut ac ir 2 M Rahimmalek Assist Prof Supervisor mrahimmalek@cc iut ac irAbstractSage Salvia sp is the most important and largest genus of Lamiaceae family This genus consists of annualand perennial plants The genus is herbaceous woody sub shrubby and often very fragrant Medicinalproperties aromatic and antioxidant of Salvia species are well known In recent years plant extracts havebeen appeared on the market as antioxidants for food industry usage The antioxidant activity of some naturalcompounds is comparable and sometimes higher than the synthetic antioxidants Many biological effects ofphenolic compounds such as antioxidant activity anti tumor and anti virus in this genus are characterized High similarity among species in this genus leads to difficult systematic identification Despite the economicimportance of this medicinal plant limited molecular studies have been done to evaluate genetic diversityamong and within Salvia species In this study SRAP sequence ralated amplified polymorphism markers targeting ORFs as functional regions in genome were used to detect the genetic diversity of five Salviaspecies S virgata S nemorosa S officinalis S cereal and S sclarea Fourteen primer combinationsampilified 265 fragments on 54 genotypes in which 255 96 were polymorphic Average PIC value was0 308 over all primer combinations The genetic distance among species ranged from 0 1263 betweenS virgata and S nemorosa to 0 5684 between S nemorosa and S sclarea Cluster analysis based onJaccard s similarity coefficient and UPGMA algorithm with cophenetic correlation coefficient of 0 9204 partly separated different species The results showed high genetic differentiation Fst 0 3368 andnegligible gene flow Nm 0 7505 among species High genetic variation among and within Salvia speciesintroduce it as a rich source of germplasm for use in breeding programs In this research after methanolicextraction of seven species S virgata S nemorosa S officinalis S cereal S sclarea S reuterana andS persica total phenolic contents was determined by means of spectrophotometry using Folin Ciocalteureagent and a wide variation has been observed among species 33 83 114 00 mg Tannic acid g dry plant Three in vitro model systems were applied to assess antioxidant activity of sage species including diphenyl picryl hydrazyl DPPH oxidation of beta carotene linoleic acid assay and Ferric reducingpower In DPPH assays and Ferric reducing power three species S virgata S officinalis S sclarea hadthe highest antioxidant activity This activity was also higher than the synthetic ones In beta carotene linoleic acid model S persica and S virgata species had the highest antioxidant activity followed byS officinalis Keywords Sage Genetic diversity SRAP marker Total phenolic content Antioxidantactivity
